目的 探讨腺病毒介入性血管内皮生长因子 (VEGF)基因导入皮下促进血管新生的可行性。方法 使用2 5G 1ml注射器将 0 .2ml含有 1× 10 9pfu( plaqueformationunit)腺病毒AdexCAhVEGF165注入小鼠腹部正中皮下。对照组注入对照腺病毒AdexCAlacZ于小鼠皮下 ,15天后处死动物 ,组织学切片随机观察皮下组织中毛细血管数量。结果 观察组皮下组织可见大量新生血管。每平方毫米含 2 2 0± 44个毛细血管 ,较对照组每平方毫米 70± 14个增加约 3倍 (t =8.4,P <0 .0 1)。结论 VEGF导入皮下可增加皮下组织毛细血管数量 ,可望用于缺血性疾病的治疗。
Objective To investigate the feasibility of subcutaneous administration of adenovirus-mediated vascular endothelial growth factor (VEGF) gene into angiogenesis. Methods 0.2 ml of AdexCAhVEGF165 containing 1 x 109 pfu (plaque forming unit) adenovirus was injected subcutaneously into the abdomen of a mouse using a 2 5 G 1 ml syringe. The control group was injected with Adenovirus AdexCAlacZ subcutaneously in mice, and the animals were sacrificed 15 days later. The number of capillaries in the subcutaneous tissue was observed by histological sections. Results A significant amount of neovascularization was observed in the subcutaneous tissue of observation group. There were 220 ± 44 capillaries per square millimeter, about 3 times more than 70 ± 14 per millimeter of the control group (t = 8.4, P <0.01). Conclusion Subcutaneous VEGF can increase the number of subcutaneous tissue capillaries, is expected to be used for the treatment of ischemic diseases.
