目的探讨转移能力不同的两种人乳腺癌细胞株MCF-7和MDA-MB-435S中Twist基因表达差异,及其与紫杉醇药物敏感性差异的相关性。方法用逆转录聚合酶链式反应(RT-PCR)方法,检测两种细胞中Twist基因的表达情况,MTT法检测两种细胞对紫杉醇的敏感性。结果 (1)RT-PCR结果显示:两种细胞中均有Twist基因表达,在MDA-MB-435S细胞中的表达高于MCF-7(P<0.05);(2)紫杉醇对MDA-MB-435S细胞的抑制作用弱于对MCF-7细胞的抑制作用(P<0.05)。结论 Twist基因在转移能力强的MDA-MB-435S细胞中表达高于转移能力中等的MCF-7细胞,同时高转移的MDA-MB-435S细胞对紫杉醇的敏感性弱于中等转移性的MCF-7细胞,且更易出现耐药。
Objective To investigate the difference of Twist gene expression in two human breast cancer cell lines MCF-7 and MDA-MB-435S with different metastatic potential and its correlation with the difference in drug sensitivity of paclitaxel. Methods The expression of Twist gene was detected by reverse transcription-polymerase chain reaction (RT-PCR) in two kinds of cells. MTT assay was used to detect the sensitivity of the two cells to paclitaxel. Results (1) RT-PCR results showed that Twist gene expression in both cell lines was higher than that in MCF-7 cells in MDA-MB-435S cells (P <0.05) The inhibitory effect of 435S cells was weaker than that of MCF-7 cells (P <0.05). Conclusions The expression of Twist gene in MCF-7 cells with high metastatic potential is higher than that in MCF-7 cells with moderate metastatic potential. Meanwhile, the MDA-MB-435S cells with high metastasis rate are less sensitive to paclitaxel than those with moderately metastatic MCF- 7 cells, and more susceptible to emergence.
