目的:观察不同运动强度训练后,大鼠肾脏血管紧张素ⅡAT1受体表达的变化,为运动对肾脏内分泌功能影响的研究提供形态学依据。方法:健康雄性SD大鼠80只分为4组,实施不同强度运动训练后,采用免疫组织化学法和计算机图像分析技术,观察分析肾脏血管紧张素ⅡAT1受体表达的分布。结果:肾脏血管紧张素ⅡAT1受体主要分布在肾小球、近曲小管和远曲小管,以远曲小管表达最为丰富。小强度运动训练后,肾脏血管紧张素ⅡAT1受体免疫反应变化不明显;中等强度训练运动后,肾脏血管紧张素ⅡAT1受体免疫反应明显减弱;大强度运动训练后,肾脏血管紧张素ⅡAT1受体免疫反应明显增强。结论:小强度运动训练对肾脏血管紧张素ⅡAT1受体表达影响不明显;中等强度运动训练使肾脏血管紧张素ⅡAT1受体表达下调,以适应运动应激;大强度运动使肾脏血管紧张素ⅡAT1受体表达上调,提示大强度运动可能有导致肾脏损害的趋势。
OBJECTIVE: To observe the changes of angiotensin Ⅱ (AT1) receptor expression in rat kidney after training with different exercise intensity, and to provide a morphological basis for the study on the effects of exercise on endocrine function of kidney. Methods: Eighty male SD rats were randomly divided into 4 groups. After training with different intensities, immunohistochemistry and computer image analysis were used to observe the distribution of renal angiotensin Ⅱ type 1 receptor. Results: The renal angiotensin Ⅱ AT1 receptors mainly distributed in the glomerulus, proximal convoluted tubule and distal convoluted tubule, and were most abundantly expressed in distal convoluted tubules. After low intensity exercise training, the renal angiotensin Ⅱ AT1 receptor immune response did not change significantly; moderate intensity training exercise, kidney angiotensin Ⅱ AT1 receptor immune response was significantly reduced; high intensity exercise training, renal angiotensin Ⅱ AT1 receptor Immune response was significantly enhanced. CONCLUSION: Small intensity exercise training has no obvious effect on the expression of angiotensin Ⅱ AT1 receptor in kidney. Middle-intensity exercise training down-regulates the expression of angiotensin Ⅱ AT1 receptor in kidney so as to adapt to exercise stress. High intensity exercise makes angiotensin Ⅱ AT1 receptor Upregulation of the body suggests that intensive exercise may have a tendency to cause kidney damage.
