目的:比较地黄不同炮制品组成的四物汤免疫作用的区别,说明生地黄和熟地黄功效的异同。方法:昆明种雌性小鼠,第1,2,3天腹腔注射(ip)环磷酰胺(CTX)80 mg.kg-1建立免疫抑制模型,按体重随机分为模型对照组,阳性香菇多糖组(0.1 g.kg-1),鲜地黄-四物汤组(20.3 g.kg-1),生地黄-四物汤组(15.8 g.kg-1),熟地黄-四物汤组(15.8 g.kg-1),去当归-四物汤组(12.0 g.kg-1),每组10只,连续灌胃(ig)7 d。取脾脏和胸腺,计算脏器指数;采集淋巴细胞样品,用流式细胞仪进行检测,观察受试药物对模型动物CD3+,CD4+,CD8+淋巴细胞及CD4+/CD8+的表达水平。结果:熟地黄-四物汤组能显著提高免疫抑制小鼠的脾脏指数(7.26±0.85)mg.g-1和胸腺指数(0.75±0.12)mg.g-1(P<0.05),同时能够极显著提高CTX造成的免疫抑制小鼠CD3+(40.68±1.24)%,CD4+(48.08±2.14)%,CD8+(29.56±1.43)%淋巴细胞(P<0.01)及CD4+/CD8+(1.62±0.36),其他组无显著性差异。结论:熟地黄-四物汤能显著提高机体T细胞及其亚群间的平衡,鲜地黄-四物汤和生地黄-四物汤作用不显著,生、熟地黄组方四物汤其对免疫功能的作用差别较大。
OBJECTIVE: To compare the difference of the immune effects of Siwu Decoction composed of different processed products of Radix Rehmanniae, and to show the similarities and differences in the efficacy of Dihuang and Rehmannia. Methods: Kunming female mice were immunosuppressed by intraperitoneal injection of cyclophosphamide (CTX) 80 mg.kg-1 on days 1, 2, and 3. The mice were randomly divided into model control group, positive lentinan group (0.1 g.kg-1), fresh rehmannia root-Siuetang group (20.3 g.kg-1), rehmannia root-Siuetang group (15.8 g.kg-1) g.kg-1), and then to Angelica sinensis-Siuetang group (12.0 g.kg-1) for 10 days. The spleen and thymus were taken out to calculate the organ index. Lymphocyte samples were collected and tested by flow cytometry. The expression levels of CD3 +, CD4 +, CD8 + lymphocytes and CD4 + / CD8 + of the model animals were observed. Results: The SPI (7.26 ± 0.85) mg.g-1 and the TH (0.75 ± 0.12) mg.g-1 of the immunosuppressed mice (P <0.05) (P <0.01), CD4 + / CD8 + (1.62 ± 0.36), CD4 + (48.08 ± 2.14)%, CD8 + (29.56 ± 1.43)% of lymphocytes in CTX-induced immunosuppressed mice Other groups no significant difference. Conclusion: Rehmanniae to Siwu Decoction can significantly improve the balance between T cell and its subpopulation, the effect of fresh Dihuang - Siwu Tang and Dihuang - Siwu Tang is not significant, The role of difference is larger.
