目的:探讨Bcl-2和MMP-1在复发子宫肌瘤组织中的表达及相关性。方法:随机选择子宫肌瘤剔除术患者100例,其中50例为首次剔除子宫肌瘤患者、50例为复发剔除子宫肌瘤患者。术前患者清晨空腹采血待测,术中取患者肌瘤组织及瘤旁正常子宫平滑肌组织待测。采用ELISA法检测患者血清、肌瘤组织及瘤旁正常子宫平滑肌组织中Bcl-2和MMP-1的蛋白含量;采用免疫组织化学方法检测首次肌瘤组织、瘤旁正常子宫平滑肌组织和复发肌瘤组织、瘤旁正常子宫平滑肌组织中Bcl-2和MMP-1的蛋白阳性表达强度。结果:与首次子宫肌瘤患者比较,复发子宫肌瘤患者血清、肌瘤组织及瘤旁正常子宫平滑肌组织中Bcl-2和MMP-1的蛋白含量均明显升高,差异有统计学意义(P<0.01)。首次瘤旁正常子宫平滑肌组织、复发瘤旁正常子宫平滑肌组织、首次肌瘤组织和复发肌瘤组织,其Bcl-2和MMP-1的蛋白含量、蛋白阳性表达强度依次增强,组间差异有统计学意义(P<0.01);且Bcl-2和MMP-1表达一致呈正相关性(r=0.032,P<0.05)。结论:Bcl-2和MMP-1在复发子宫肌瘤患者血清及瘤组织的高表达,是子宫肌瘤的复发因素,其机制有待于进一步研究。
Objective: To investigate the expression and correlation of Bcl-2 and MMP-1 in recurrent uterine fibroids. Methods: 100 cases of myomectomy were randomly selected, of which 50 cases were patients with uterine myoma removed for the first time and 50 cases were patients with recurrence and rejection of uterine fibroids. Preoperative fasting blood samples were taken early in the morning, intraoperative take myoma and para-normal uterine smooth muscle tissue to be tested. The protein levels of Bcl-2 and MMP-1 in serum, fibroids and normal uterine smooth muscle tissues of patients were detected by ELISA. The primary myometrium, normal uterine smooth muscle and recurrence myoma were detected by immunohistochemistry The positive expression of Bcl-2 and MMP-1 protein in normal uterine smooth muscle tissue of tumor tissue was detected by immunohistochemistry. Results: The protein levels of Bcl-2 and MMP-1 in serum, fibroid tissue and normal uterine smooth muscle tissue of patients with recurrent uterine fibroids were significantly higher than those of the first patients with uterine fibroids (P <0.01). The protein level and protein expression of Bcl-2 and MMP-1 in the normal para-normal uterine smooth muscle tissue, normal uterine tissue next to tumor recurrence, first myoma and recurrence myoma were increased (P <0.01). The expressions of Bcl-2 and MMP-1 were positively correlated (r = 0.032, P <0.05). Conclusion: The high expression of Bcl-2 and MMP-1 in the serum and tumor tissues of patients with recurrent uterine fibroids is a recurrence factor of uterine fibroids, and its mechanism remains to be further studied.
