以3种基因型的小型西瓜无菌苗为试材,采用体外培养的方法,研究了外植体类型、激素组合、继代生根培养对离体器官再生的影响。结果表明:以子叶节为外植体的不定芽再生效果好于子叶;不同基因型最适的激素组合不同,W1的最适诱导培养基是MS+1.0mg/L 6-BA+0.5mg/L NAA,再生频率为20.00%,W2的最适诱导培养基是MS+3.0mg/L 6-BA+0.5mg/L NAA,再生频率为30.00%,W3的最适诱导培养基是MS+2.0mg/L 6-BA+0.5mg/L NAA,再生频率为24.00%;最佳生根培养基为MS+NAA 0.5mg/L。
Using three genotypes of small watermelon sterile seedlings as test materials, the effects of explant types, hormone combinations and subculture on isolated organ regeneration were studied in vitro. The results showed that adventitious shoot regeneration with cotyledonary node as explant was better than that of cotyledon. The most suitable medium for inducing W1 was MS + 1.0mg / L 6-BA + 0.5mg / L NAA, the regeneration frequency was 20.00%. The optimal induction medium for W2 was MS + 3.0mg / L 6-BA + 0.5mg / L NAA, the regeneration frequency was 30.00%, and the optimal induction medium for W3 was MS + 2.0 mg / L 6-BA + 0.5mg / L NAA, the regeneration frequency was 24.00%. The best rooting medium was MS + NAA 0.5mg / L.
