目的探讨牙龈卟啉单胞菌(Porphyromonas gingivalis,Pg)对骨细胞凋亡的诱导作用,及其对骨质破坏的影响。方法 2009年8-12月于昆明医学院口腔实验室,选取昆明小鼠60只,随机分为实验组和对照组,每组30只。于实验组小鼠头骨中线两耳之间的部位注射一定浓度的Pg细菌悬液;对照组动物在同样条件下注射等量的0.01mol/L PBS液。分别于注射后当即及第1、3、5、8、12天处死,保留整个头颅骨进行固定、脱钙和包埋,原位末端标记(TUNEL)法观察骨细胞的凋亡状况,并计算细胞凋亡率。结果实验组在注射后第1天,细胞凋亡率较注射前显著增加,第5天达峰值[(35.64%±3.69)%],第8、12天的细胞凋亡率有所下降;与对照组相比,差异均有统计学意义(P<0.05)。结论 Pg具有诱导骨细胞凋亡的作用,提示大量骨细胞凋亡可能是牙周炎骨组织破坏产生而重建愈合相对不足的原因之一。
Objective To investigate the effect of Porphyromonas gingivalis (Pg) on the apoptosis of osteoblasts and its effect on bone destruction. Methods Eighty Kunming mice were randomly divided into experimental group and control group at the oral laboratory of Kunming Medical College from August to December in 2009, with 30 in each group. In the experimental group mice skull midline between the two parts of the injection of a certain concentration of Pg bacterial suspension; the control group animals under the same conditions the same amount of 0.01mol / L PBS solution. The rats were killed immediately after injection and on the 1st, 3rd, 5th, 8th and 12th days respectively. The entire skull was preserved for fixation, decalcification and embedding. TUNEL method was used to observe the apoptosis of osteocytes. Rate of apoptosis. Results On the first day after injection, the apoptotic rate of the experimental group was significantly higher than that before injection, reaching the peak on the fifth day [(35.64% ± 3.69)%], and the apoptosis rate decreased on the 8th and 12th days. Compared with the control group, the differences were statistically significant (P <0.05). Conclusions Pg can induce the apoptosis of osteoblasts, suggesting that a large number of apoptosis of osteocytes may be one of the causes of bone tissue destruction and reconstructive healing in periodontitis.
