熊胆对自发性高血压大鼠肠系膜动脉血管结构和舒张功能的影响

目的观察熊胆对自发性高血压大鼠(SHR)血压、肠系膜动脉血管结构和舒张功能的影响。方法 12周龄雄性SHR 16只随机分为:SHR熊胆治疗组[SHR-X,n=8,800mg/(kg·d)]和SHR对照组(SHR,n=8),并选取对应周龄雄性Wistar Kyoto大鼠作为正常血压对照组(WKY,n=8),灌胃8周。采用无创尾袖法测量大鼠尾动脉收缩压。应用计算机图像分析系统计算HE染色后的血管管壁面积/管腔面积(W/L)、管壁厚度/管腔半径(WT/LR)。采用PowerLab生物信息采集系统检测离体肠系膜动脉对不同浓度血管活性物质的舒张反应。结果熊胆治疗4和8周末,SHR-X组的血压明显低于未治疗的SHR组[收缩压,4周:SHR-X(176.4±11.4)比SHR(200.3±13.9)mm Hg;8周:SHR-X(169.4±12.0)比SHR(189.9±10.1)mm Hg;均P<0.01]。熊胆治疗8周末,SHR-X组的肠系膜动脉W/L及WT/LR明显低于未治疗的SHR(W/L:SHR-X 0.53±0.09比SHR 1.82±0.96;WT/LR:SHR-X 0.24±0.08比SHR 0.53±0.29,均P<0.01),与WKY组比较差异无统计学意义(P>0.05);SHR-X的肠系膜动脉内皮依赖性血管舒张功能明显高于未治疗SHR[最大舒张率:SHR-X(59.29±15.15)%比SHR(20.69±6.31)%,舒张反应敏感性(pD2):SHR-X 8.24±0.32比SHR 5.82±0.23;均P<0.01],非内皮依赖性血管舒张功能也明显高于SHR组[最大舒张率:SHR-X(96.37±1.87)%比SHR(29.04±4.56)%;pD2:SHR-X 7.79±0.15比SHR 5.31±0.14;均P<0.01]。结论熊胆治疗可轻度降低SHR的血压,明显改善肠系膜动脉血管结构和舒张功能。 Objective To observe the effects of bear bile on the blood pressure, the structure of mesenteric artery and the diastolic function in spontaneously hypertensive rats (SHR). Methods SHR-X (n = 8, 800 mg / (kg · d)) and SHR control group (SHR, n = 8) were randomly divided into two groups at random. Male Wistar Kyoto rats were used as a normal blood pressure control group (WKY, n = 8) for 8 weeks. The tail artery systolic pressure was measured by noninvasive tail cuff method. The area of ​​the vascular wall / lumen area (W / L) and the thickness of the lumen / lumen radius (WT / LR) after HE staining were calculated by computer image analysis system. Results After 4 and 8 weeks of treatment, the blood pressure of SHR-X group was significantly lower than that of untreated SHR group [4 weeks: SHR-X (176.4 ± 11.4) vs SHR (200.3 ± 13.9) mm Hg; : SHR-X (169.4 ± 12.0) vs SHR (189.9 ± 10.1) mm Hg; all P <0.01]. The W / L and WT / LR of mesenteric artery in SHR-X group were significantly lower than those in untreated SHR-X group (W / L: 0.53 ± 0.09 vs 1.82 ± 0.96 vs SHR; X 0.24 ± 0.08 vs SHR 0.53 ± 0.29, all P <0.01), but there was no significant difference between the two groups (P> 0.05). The function of SHR-X in mesenteric artery was significantly higher than that of untreated SHR [ The maximum relaxation rate of SHR-X was (59.29 ± 15.15)% compared with SHR (20.69 ± 6.31)%, pD2: SHR-X 8.24 ± 0.32 vs SHR 5.82 ± 0.23; all P <0.01] Dependent vasodilation was also significantly higher than that of SHR group [maximal diastolic rate: SHR-X (96.37 ± 1.87)% vs SHR (29.04 ± 4.56)%; pD2: SHR-X 7.79 ± 0.15 vs SHR 5.31 ± 0.14 <0.01]. Conclusion Bear gall can reduce the blood pressure of SHR slightly and obviously improve the structure of mesenteric artery and the diastolic function.
