目的:探讨芪麝颈康丸对大鼠退变颈椎间盘碱性成纤维细胞生长因子(bFGF)及胰岛素样生长因子(IGF-Ⅰ)mRNA 表达的调节作用。方法:建立动静力失衡性颈椎间盘退变模型,逆转录多聚酶链式反应方法检测椎间盘组织内 bFGF、IGF-ImRNA 表达水平。结果:模型组 bFGF mRNA 表达明显,与正常对照组比较差异有显著性(P<0.01);与模型组比较,芪麝颈康丸、活血通络方、芬必得都可以下调 bFGF,尤以芪麝颈康丸下调明显(P<0.01)。模型组 IGF-Ⅰ mRNA 表达明显减弱,与正常对照组比较差异有显著性(P<0.01);芪麝颈康丸可以上调 IGF-ⅠmRNA(P<0.01),活血通络方、芬必得作用不明显。结论:芪麝颈康丸可以在下调椎间盘组织内 bFGF mRNA,上调 IGF-Ⅰ mRNA 的基因表达。
OBJECTIVE: To investigate the regulation of basic cervical fibroblast growth factor (bFGF) and insulin-like growth factor (IGF-I) mRNA expression in rats with degenerative cervical intervertebral disc. METHODS: An imbalanced model of cervical intervertebral disc degeneration was established. Reverse transcription-polymerase chain reaction was used to detect the expression of bFGF and IGF-I mRNA in intervertebral disc tissue. Results: The expression of bFGF mRNA in the model group was significantly higher than that of the normal control group (P<0.01). Compared with the model group, both the Huojiankang pill, Huoxuetongluo decoction, and Fenbid may all lower bFGF, especially in the model group. There was significant down-regulation of Qijingkang Pill (P<0.01). The expression of IGF-I mRNA in the model group was significantly attenuated, and there was a significant difference compared with the normal control group (P<0.01). The IGF-I mRNA could be up-regulated by Shujingkang Pill (P<0.01). The effect of Huoxue Tongluo Fang and Fenbid would not be effective. obvious. Conclusions: Zishangkang Pill can down-regulate bFGF mRNA in intervertebral disc tissue and up-regulate IGF-I mRNA expression.
