目的:旨在探讨黄芩素(BAI)对血管生成的影响,及血管内皮生长因子(VEGF)对多发性骨髓瘤细胞系RPMI-8226细胞生长的影响。方法:体外培养RPMI-8226细胞系,不同浓度BAI处理此细胞系,用Annexin-V/PI双标流式细胞术检测BAI对RPMI-8226细胞系凋亡的作用,应用RT-PCR检测黄芩素对RPMI-8226细胞VEGF表达的改变。结果:经不同浓度BAI(0,25,50,100μg/L)处理后的RPMI-8226细胞,随着BAI浓度的增加,细胞的早期凋亡及总凋亡率增加,以早期凋亡更明显;随着BAI(0,50,100μg/L)浓度增加,VEGF基因的表达呈下降趋势。结论:在一定浓度范围内,黄芩素可促进RPMI-8226细胞发生凋亡,且呈剂量依赖性,该过程与降低骨髓瘤细胞VEGF表达,抑制血管内皮生长作用有关。
Objective: To investigate the effect of baicalein (BAI) on angiogenesis and the effect of vascular endothelial growth factor (VEGF) on the growth of multiple myeloma RPMI-8226 cells. Methods: The RPMI-8226 cell line was cultured in vitro and treated with BAI at different concentrations. The apoptosis of RPMI-8226 cell line was detected by Annexin-V / PI dual-flow cytometry. The expression of baicalein Changes of VEGF expression in RPMI-8226 cells. Results: After treated with different concentrations of BAI (0, 25, 50 and 100 μg / L), the early apoptosis and total apoptosis of cells were increased with the increase of BAI concentration. With the increase of BAI concentration (0,50,100μg / L), the expression of VEGF gene tended to decrease. CONCLUSION: Baicalein can promote the apoptosis of RPMI-8226 cells in a dose-dependent manner in a certain concentration range, which is related to the decrease of VEGF expression and the inhibition of vascular endothelial growth in myeloma cells.
