目的观察急性心肌缺血大鼠心肌组织内孤啡肽(orphanin FQ,OFQ)的表达变化,探讨孤啡肽在急性心肌缺血中的作用。方法 54只260~280g健康成年雄性SD大鼠,按随机数字表法分为3组:①动物对照组(C组);②手术对照组(Sham组);③结扎冠状动脉组(CAO组)。采用免疫组织化学染色(SABC法),观察冠脉结扎大鼠不同时点心肌组织内OFQ的表达变化,以平均光密度为指标进行半定量分析。同时记录相应时点心电图波形,并对不同时点P波、T波振幅和ST段偏移量进行统计学分析。结果免疫组化切片CAO后各时点心肌缺血区OFQ的水平较对照组显著升高(P<0.05),在CAO3h达高峰(0.663±0.021)(P<0.01)。与对照组心电图相比,CAO组不同时点P波、T波振幅和ST段偏移量差别有统计学意义(P<0.05)。结论急性心肌缺血时OFQ表达显著增多,可能起到心肌保护作用。
Objective To investigate the expression of orphanin FQ (OFQ) in myocardium of acute myocardial ischemia rats and to explore the role of orphanin in acute myocardial ischemia. Methods 54 adult healthy male Sprague-Dawley rats weighing 260-280 g were randomly divided into three groups according to the random number table: ① animal control group (C group); ② surgery control group (Sham group); ③ coronary artery ligation group (CAO group) . Immunohistochemical staining (SABC method) was used to observe the changes of OFQ expression in myocardial tissue at different time points after coronary ligation, and the semi-quantitative analysis was performed with the average optical density as an index. At the same time, the waveform of electrocardiogram was recorded at the same time, and the P wave, T wave amplitude and ST segment offset at different time points were statistically analyzed. Results The level of OFQ in myocardial ischemic area at each time point after CAO was significantly higher than that in control group (P <0.05) and peaked at CAO3 (0.663 ± 0.021) (P <0.01). Compared with control group, P wave, T wave amplitude and ST segment offset at different time points in CAO group were significantly different (P <0.05). Conclusion OFQ expression is significantly increased in acute myocardial ischemia, which may play a role in myocardial protection.
