目的探讨参麦注射液在单侧输尿管梗阻(UUO)大鼠肾间质纤维化进程中的可能作用。方法成年SD大鼠54只,随机分为假手术组、模型组和参麦治疗组。假手术组仅完成开腹过程,不结扎输尿管;模型组和参麦治疗组行左侧输尿管结扎术,术后参麦治疗组每天腹腔注射参麦注射液3ml/(kg.d),假手术组与模型组则每天腹腔注射等量生理盐水。各组分别于实验的d7、d14、d21处死动物6只,取左肾组织行HE染色和α-平滑肌肌动蛋白(α-SMA)免疫组化检查,并测定梗阻侧肾组织中SOD和MDA含量。结果与模型组比较,参麦治疗组肾小管间质病理改变明显减轻,肾间质α-SMA表达减少;肾组织中SOD活性增加,MDA含量下降。结论参麦注射液可通过减少氧化应激,减少肾间质α-SMA表达而抑制肾间质纤维化进程。
Objective To investigate the possible role of Shenmai injection in the process of renal interstitial fibrosis in unilateral ureteral obstruction (UUO) rats. Methods Fifty-four adult SD rats were randomly divided into sham operation group, model group and Shenmai treatment group. The sham-operation group only finished the laparotomy and did not ligate the ureter. The model group and the Shenmai treatment group underwent the left ureteral ligation. After the treatment, the Shenmai treatment group was given intraperitoneal injection of Shenmai injection 3ml / (kg.d) Group and model group were injected intraperitoneally with normal saline every day. Six rabbits were sacrificed on d7, d14 and d21 of the experiment, HE staining and α-smooth muscle actin (α-SMA) immunohistochemistry were performed on the left kidney and SOD and MDA content. Results Compared with the model group, the pathological changes of tubulointerstitial of Shenmai treatment group were obviously alleviated, and the expression of α-SMA in renal interstitium was decreased. The activity of SOD in renal tissue was increased and the content of MDA was decreased. Conclusion Shenmai injection can inhibit the process of renal interstitial fibrosis by reducing oxidative stress and decreasing α-SMA expression in renal interstitium.
