生长激素刺激培养新生大鼠心肌细胞,用含MBP凝胶分离法测定细胞外信号调节酶(ERK)活性。用WesternBlooting法检测磷酸化的p38MAPK,观察心肌细胞内MAPK超家族活性变化,探讨心肌细胞GH信号传导途径。结果发现:生长激素激活心肌细胞ERKs呈时间和浓度依赖性关系。而对p38MAPK无磷酸化激活作用。表明心肌生长激素信号传导通过MAPK超家族成员ERK途径,而与p38MAPK途径无关。生长激素诱导的心肌细胞ERK激活,可能是生长激素心脏效应的重要机制之一。
Growth hormone stimulates the culture of neonatal rat cardiomyocytes and the activity of extracellular signal-regulated kinase (ERK) is determined by MBP-containing gel separation. The phosphorylated p38MAPK was detected by Western Blotting, and the changes of MAPK superfamily activity in cardiomyocytes were observed to explore the GH signal transduction pathways of cardiomyocytes. The results showed that growth hormone activated cardiomyocytes ERKs in a time-and concentration-dependent manner. While p38MAPK no phosphorylation activation. Suggesting that cardiac myosin signaling through the ERK pathway of the MAPK superfamily member has nothing to do with the p38 MAPK pathway. Growth hormone-induced ERK activation of cardiac myocytes may be one of the important mechanisms of the growth hormone cardiac effects.
