温中益气胶囊对幽门结扎型胃溃疡大鼠的治疗作用

目的:研究温中益气胶囊对幽门结扎型胃溃疡大鼠的治疗作用以及对小鼠的抗炎、镇痛作用。方法:采用幽门结扎法复制大鼠胃溃疡模型。取清洁级健康SD大鼠60只,雌雄各半,随机分成空白组,模型组,阳性对照药三九胃泰颗粒组(3.69 g·kg-1),温中益气胶囊高、中、低剂量组(2.21,1.11,0.55 g·kg-1)。每天ig给药1次,连续给药7 d,模型组和空白组给予等量蒸馏水。第5天给药后各组动物禁食不禁水48 h,末次给药后2 h麻醉,进行幽门结扎手术,缝合后直肠给药1次,禁食水。18 h后麻醉大鼠,颈总动脉取血。取全胃观察大鼠胃溃疡面积和溃疡指数,胃组织匀浆测定丙二醛(MDA)、超氧化物歧化酶(SOD)、H+-K+-ATP酶、生长抑素(SS)含量。采用二甲苯致小鼠耳廓肿胀和热板刺激法进行抗炎和镇痛实验,测定小鼠耳肿胀度、抑制率及痛阈值的大小。分别取清洁级小鼠50只,随机分成空白组、阳性对照药三九胃泰颗粒组(5.19 g·kg-1)、温中益气胶囊高、中、低剂量组(3.12,1.56,0.78 g·kg-1)。结果:胃溃疡实验中,与空白组比较,模型组H+-K+-ATP酶活性和MDA含量显著增加(P<0.01),SS和SOD活性显著降低(P<0.05,P<0.01);与模型组比较,温中益气胶囊能显著减小大鼠胃溃疡面积和溃疡指数,降低H+-K+-ATP酶的活性和MDA含量并增加胃组织中SS和SOD水平(P<0.01,P<0.05)。抗炎、镇痛实验中,与空白组比较,温中益气胶囊能降低二甲苯致小鼠耳廓肿胀度,提高小鼠痛阈值(P<0.01或P<0.05)。结论:温中益气胶囊可能通过降低H+-K+-ATP酶活性和MDA含量、增强SOD活性、增加SS的含量来实现抗幽门结扎型胃溃疡作用,并具有明显的抗炎、镇痛作用。 Objective: To study the therapeutic effect of Wenzhongyiqi capsule on pylorus-ligated gastric ulcer rats and its anti-inflammatory and analgesic effects on mice. Methods: The rat model of gastric ulcer was established by pylorus ligation. Sixty healthy male SD rats were randomly divided into blank group, model group and positive control drug Sanjiuweitai Granule group (3.69 g · kg-1) .Wuhan Zhongyi Capsule high, middle and low dose group (2.21,1.11,0.55 g · kg -1). Administered ig 1 times a day, continuous administration of 7 d, the model group and the blank group were given the same amount of distilled water. After the 5th day, the animals in each group were fasted for 48 hours and were anesthetized 2 hours after the last administration. The patients underwent pyloric ligation surgery, rectally administered once a day after suture, and fasting water. After 18 h, rats were anesthetized and the common carotid artery was taken for blood. Gastric ulcer area and ulcer index were taken from the whole stomach. The contents of malondialdehyde (MDA), superoxide dismutase (SOD), H + -K + -ATPase and somatostatin (SS) in gastric homogenate were measured. The mice were subjected to anti-inflammatory and analgesic experiments using mouse ear auricle swelling and hot-plate stimulation, and the ear swelling, the rate of inhibition and the threshold of pain were measured. Fifty clean mice were randomly divided into three groups: sham group (5.19 g · kg-1), high-dose Zhongwenqi capsule (3.12,1.56,0.78 g · kg-1). Results: Compared with the blank group, the activity of H + -K + -ATPase and the content of MDA in the model group were significantly increased (P <0.01) and the activities of SS and SOD were significantly decreased (P <0.05, P <0.01) Compared with the control group, Wenzhongyiqi capsule could significantly reduce the gastric ulcer area and ulcer index, reduce the activity of H + -K + -ATPase and the content of MDA and increase the levels of SS and SOD in gastric tissue (P <0.01, P <0.05). Compared with the blank group, Wen Zhong Yi Qi Capsule can reduce the auricle swelling caused by xylene and increase the pain threshold (P <0.01 or P <0.05) in the anti-inflammatory and analgesic experiments. Conclusion: Wen Zhong Yi Qi Capsule may have anti-pyloric ligation-induced gastric ulcer effect by reducing H + -K + -ATPase activity and MDA content, increasing SOD activity and increasing SS content, and has obvious anti-inflammatory and analgesic effects.
