目的研究齐墩果酸(oleanolic acid,OA)对蛛网膜下腔出血(subarachnoid hemorrhage,SAH)后早期脑损伤的影响,并探讨其潜在的作用机制。方法 39只大鼠随机分为假手术组、模型组、OA治疗组(OA组,20 mg·kg-1),每组13只。分别对各组进行体质量测量,神经功能评分,伊文思蓝法检测血脑屏障的通透性,并用Western blot法检测大鼠脑皮质中NF-κB/ICAM-1信号通路蛋白的表达情况。结果与模型组比较,OA组能显著增加大鼠体质量和神经功能评分,显著降低脑部伊文思蓝通透率和NF-κB/ICAM-1信号通路蛋白的表达水平,差异均具有统计学意义(P<0.01或P<0.05)。结论齐墩果酸能够明显减轻大鼠SAH后早期脑损伤,有可能是通过减少Akt/NF-κB/ICAM-1信号通路的炎症因子发挥作用。
Objective To investigate the effect of oleanolic acid (OA) on early brain injury after subarachnoid hemorrhage (SAH) and to explore its underlying mechanism. Methods Thirty-nine rats were randomly divided into sham-operation group, model group and OA treatment group (OA group, 20 mg · kg-1), with 13 rats in each group. Body mass measurement, neurological function score, Evans blue method were used to detect the permeability of the blood-brain barrier in each group. Western blot was used to detect the protein expression of NF-κB / ICAM-1 signaling pathway in rat cerebral cortex. Results Compared with the model group, the OA group could significantly increase the body weight and neurological function score, significantly reduce the brain Evans blue permeability and NF-κB / ICAM-1 signaling pathway protein expression, the differences were statistically significant Significance (P <0.01 or P <0.05). Conclusion Oleanolic acid can significantly reduce the early brain injury after SAH in rats, which may play an important role by reducing the inflammatory factors of Akt / NF-κB / ICAM-1 signaling pathway.
