目的:探讨三七总苷对肝癌细胞QGY-7703中mcl-1L基因与Bak基因表达水平的影响。方法:采用不同浓度三七总苷诱导肝癌细胞,通过实时荧光定量PCR检测mcl-1L基因与Bak基因的表达情况,同时采用流式细胞仪检测细胞凋亡的改变。结果:三七总苷作用后,肝癌细胞mcl-1L基因的表达受到明显抑制,而Bak基因的表达则出现明显上调,mcl-1L/Bak呈上升趋势,细胞增殖受到抑制。结论:三七总苷可能通过影响mcl-1L/Bak表达水平影响细胞凋亡。
Objective: To investigate the effects of Panax Notoginseng glycosides on the expression of mcl-1L and Bak in hepatocellular carcinoma cell line QGY-7703. Methods: Hepatic cancer cells were induced by different concentrations of Panax notoginseng saponins. The expression of mcl-1L gene and Bak gene were detected by real-time fluorescence quantitative PCR. Apoptosis was detected by flow cytometry. Results: After treated with Panax notoginseng, the expression of mcl-1L gene was significantly inhibited in hepatocellular carcinoma cells, whereas the expression of Bak gene was significantly up-regulated. The expression of mcl-1L / Bak was up-regulated and the cell proliferation was inhibited. Conclusion: Panax notoginseng glycosides may affect cell apoptosis by affecting the expression of mcl-1L / Bak.
