目的:观察补脾益气方对脾虚哮喘大鼠模型肺组织Toll样受体4(TLR4)表达的影响,探讨脾虚对哮喘大鼠气道炎症影响机制及补脾益气方对其的调控作用。方法:36只健康雄性Wistar大鼠,随机分为3组:对照组、脾虚哮喘组、脾虚哮喘治疗组,每组12只。采用RT-PCR检测大鼠肺组织TLR4 mRNA表达水平;采用免疫组化检测大鼠肺组织TLR4的蛋白表达水平。结果:和对照组比较,脾虚哮喘组大鼠肺组织TLR4 mRNA和蛋白表达水平显著增高(P<0.01);和脾虚哮喘组比较,脾虚哮喘治疗组大鼠肺组织TLR4 mRNA和蛋白表达水平显著降低(P<0.01)。结论:脾虚哮喘组大鼠肺组织中TLR4 mRNA及蛋白表达水平显著增高,补脾益气方能够降低脾虚哮喘大鼠肺组织中TLR4 mRNA和蛋白表达水平。
OBJECTIVE: To observe the effect of Bupi Yiqi Prescription on the expression of Toll-like Receptor 4 (TLR4) in the lung tissue of asthmatic rats with spleen-deficiency syndrome and to explore the mechanism of the effect of Spleen Deficiency on airway inflammation in asthmatic rats and its regulation by BuBu Yi Qi Fang . Methods: Thirty - six healthy male Wistar rats were randomly divided into three groups: control group, spleen - asthma group and spleen - asthma group, 12 in each group. The expression of TLR4 mRNA in lung tissue was detected by RT-PCR. The protein expression of TLR4 in lung tissue was detected by immunohistochemistry. Results: Compared with the control group, the expression of TLR4 mRNA and protein in the lung tissue of asthmatic rats were significantly increased (P <0.01). Compared with the asthmatic rats, the expression of TLR4 mRNA and protein in the lung tissue of asthmatic rats were significantly decreased (P <0.01). Conclusion: The TLR4 mRNA and protein expression levels in the lung tissue of asthmatic rats with spleen asthma were significantly increased. BuBu Yi Qi Fang could reduce the expression of TLR4 mRNA and protein in the lung tissue of rats with spleen asthma.
