目的观察利血平致大鼠肾上腺髓质增生(AMH)的组织学及超微结构改变。方法雄性Wistar大鼠75只,随机分为A、B、C三组各25只,A组皮下注射利血平每天0.4 mg/kg体重和0.9%生理盐水每天0.4 ml/kg体重,B组皮下注射利血平每天0.8 mg/kg体重,C组皮下注射0.9%生理盐水每天0.8 ml/kg体重,每日1次,持续2个月后停药。分别于停药后2周、2个月、6个月时处死大鼠,取出肾上腺,作光镜及电镜观察,测肾上腺髓质百分数及AMH阳性率。结果在各停药时段,A、B两组肾上腺髓质光镜及电镜观察均显示髓质细胞呈增殖活跃状态,且大鼠髓质百分数与C组比显著升高(P<0.05);停药2、6月时,A、B、C各组大鼠肾上腺髓质百分数与停药2周比较差异均无统计学意义(P>0.05);A、B组之间AMH阳性率差异无统计学意义(P>0.05)。结论利血平所致大鼠肾上腺髓质增生性改变明显、稳定,大鼠AMH与人类AMH在形态学改变方面有较大的相似性。
Objective To observe the histological and ultrastructural changes of rat adrenal hyperplasia (AMH) induced by reserpine. Methods A total of 75 male Wistar rats were randomly divided into three groups (A, B and C): 25 rats in group A, 0.4 mg / kg body weight in group A and 0.4 ml / kg body weight in normal saline 0.9% 0.8 mg / kg body weight per day for reserpine and 0.8 ml / kg body weight 0.9% saline for group C subcutaneously once daily for 2 months. The rats were sacrificed at 2 weeks, 2 months and 6 months after drug withdrawal, respectively. The adrenal glands were removed, and the percentage of adrenal medulla and the percentage of AMH were measured by light microscope and electron microscope. Results During the withdrawal period, the adrenal medulla of group A and group B showed the proliferative activity of medullary cells and the percentage of medulla was significantly higher than that of group C (P <0.05) There was no significant difference in the percentage of adrenal medulla between groups A, B and C at 2 and 6 months after drug withdrawal (P> 0.05), but there was no statistical difference in the positive rate of AMH between groups A and B Significance (P> 0.05). CONCLUSIONS: Reserpine-induced adrenal medullary hyperplasia changes significantly and steadily. There is a great similarity between AMH and human AMH in morphological changes.
