目的通过观察固本活血壮骨颗粒对去卵巢大鼠成骨细胞骨形态形成蛋白因子II(BMP-2)表达的影响,探讨固本活血壮骨颗粒防治骨质疏松症的微观机制;方法取3月龄的雌性SD大鼠60只,随机分为:假手术组、模型组、仙灵骨葆胶囊组、尼尔雌醇组、固本活血壮骨颗粒小剂量组(简称GBHX小剂量组)、大剂量组(简称GBHX大剂量组)。除假手术组仅切除双侧卵巢附近的相当于卵巢大小的小块脂肪,其余各组均切除双侧卵巢。术后一周开始给药,灌胃90 d后,取右侧股骨近段,免疫组化法测定股骨成骨细胞BMP-2表达。结果 模型组BMP-2表达明显低于假手术组(P<0.01),固本活血壮骨颗粒大剂量组BMP-2表达明显高于模型组(P<0.01),且明显高其他治疗组。结论 固本活血壮骨颗粒可通过刺激成骨细胞BMP-2表达促进骨形成,且有剂量依赖关系。
Objective To observe the effect of Gubenhuoxuezhuanggu granules on the expression of bone morphogenetic protein factor II (BMP-2) in ovariectomized rats and explore the mechanism of Guben Huoxuezhuanggu granules in prevention and treatment of osteoporosis.Methods Sixty-three-month-old female Sprague Dawley rats were randomly divided into four groups: sham operation group, model group, Xianling Gubao Capsule group, nilestriol group, Guben Huoxuezhuanggu granules low dose group ), High-dose group (GBHX high-dose group). In addition to sham operation group only resected ovarian size of small fat near the bilateral ovaries, the other groups were removed bilateral ovaries. One week after the operation, the proximal femur was taken 90 days after gavage, and BMP-2 expression in femoral osteoblast was detected by immunohistochemistry. Results The expression of BMP-2 in model group was significantly lower than that in sham operation group (P <0.01). The BMP-2 expression of Gubeng Huoxuogu Granules group was significantly higher than that in model group (P <0.01) and obviously higher in other treatment groups. Conclusion Gubenhuoxuezhuanggu granules can promote bone formation by stimulating osteoblast BMP-2 expression in a dose-dependent manner.
