目的:观察黄连解毒汤70%醇提物对MDR模型小鼠S180肉瘤细胞凋亡因子的影响,探讨其逆转多药耐药作用的分子机制。方法:模拟临床PFC方案,建立小鼠S180肉瘤细胞多药耐药在体模型,同时给予黄连解毒汤70%醇提物灌胃10d,流式细胞仪荧光检测观察细胞凋亡因子Fas、Trail、细胞黏附因子CD54等指标。结果:黄连解毒汤70%醇提物增加耐药细胞凋亡因子Fas、Trail表达率,促进耐药S180细胞的凋亡,降低细胞黏附因子CD54的表达。结论:黄连解毒汤70%醇提物促进耐药细胞凋亡因子的高表达,是其逆转肿瘤多药耐药的途径之一。
Objective: To observe the effect of 70% alcohol extract of Huanglian Jiedu Decoction on the apoptosis of S180 sarcoma cells in MDR model mice and to explore the molecular mechanism of reversing multidrug resistance. Methods: A multi-drug resistance model of S180 sarcoma cells in mice was established by simulating a clinical PFC protocol. At the same time, a 70% alcohol extract of Huanglian Jiedu Decoction was given for 10 days. Flow cytometry was used to observe the apoptosis factors Fas and Trail. Cell adhesion factor CD54 and other indicators. Results: The 70% alcohol extract of Huanglian Jiedu Decoction increased the expression rate of Fas and Trail apoptotic factors in drug-resistant cells, promoted the apoptosis of drug-resistant S180 cells, and decreased the expression of cell adhesion factor CD54. Conclusion: The 70% alcohol extract of Huanglian Jiedu Decoction promotes the high expression of apoptotic factors in drug-resistant cells, which is one of the ways to reverse the multidrug resistance of tumor cells.
