从毛竹全长cDNA文库中分离到1个细胞色素P450基因,命名为PheCYP-1。相似性分析表明,其编码蛋白与拟南芥CYP706A亚家族蛋白相似性较高(42%~46%),属于CYP706家族;实时定量PCR结果表明,PheCYP-1在正在伸长的笋中表达丰度最高;将PheCYP-1构建到pBI121的多克隆位点,在拟南芥中过量表达,转基因拟南芥的次生木质部导管壁厚度显著增加,表明PheCYP-1基因可能与毛竹次生木质部的细胞壁发育有关。
One Cytochrome P450 gene was isolated from the full - length cDNA library of Phyllostachys pubescens and named PheCYP - 1. Similarity analysis showed that the protein encoded by CYP706A was highly similar (42% -46%) to CYP706 family, and the real-time quantitative PCR results showed that PheCYP-1 expressed abundance in growing shoots PheCYP-1 was constructed to multiple cloning sites in pBI121 and overexpressed in Arabidopsis thaliana, and the secondary xylem vessel wall thickness in transgenic Arabidopsis thaliana was significantly increased, indicating that PheCYP-1 may be associated with the secondary xylem Cell wall development related.
