本试验旨在探讨不同补饲日龄对湖羊羔羊瘤胃形态发育以及瘤胃表皮生长相关基因表达的影响。本试验采用两因素试验设计,设补饲日龄和羔羊日龄两个因子。选择初生重((3.41±0.27)kg)接近的78只双羔湖羊,0日龄时随机屠宰6只,按照同质性原则将剩余72只湖羊分为7d补饲组((3.36±0.15)kg)和42d补饲组((3.44±0.44)kg)。7d补饲组的羔羊从7日龄补饲开食料1,42d补饲组的羔羊42日龄补饲开食料1,两组羔羊56日龄断奶,60日龄时开始逐步换开食料2,过渡期10d。分别在14、28、42、56、70和84日龄从两组中各随机挑选6只羔羊进行屠宰,迅速采集瘤胃腹囊,采用Real-time qPCR方法分析瘤胃表皮生长相关基因表达量的发育性变化,并制备切片以观察瘤胃发育。试验结果表明,7d补饲组羔羊瘤胃乳头宽度显著高于42d补饲组羔羊(P<0.05),但瘤胃肌层厚度显著低于42d补饲组羔羊(P<0.05)。7d补饲组羔羊瘤胃上皮IGFBP3表达量显著低于42d补饲组羔羊(P<0.05),瘤胃上皮IGFBP5表达量显著高于42d补饲组羔羊(P<0.05)。7d补饲组羔羊瘤胃乳头长度和宽度与瘤胃上皮TGFβ1和IGFBP3表达量呈显著正相关(R=0.507,P<0.001;R=0.444,P<0.001;R=0.465,P<0.001;R=0.299,P=0.011),与瘤胃上皮IGFBP6表达量呈显著负相关(R=-0.443,P<0.001;R=-0.447,P<0.001);42d补饲组羔羊瘤胃乳头长度与IGFBP5表达量呈显著正相关(R=0.227,P=0.018),瘤胃乳头宽度与瘤胃上皮IGFBP3和IGFBP5表达量呈显著正相关(R=0.338,P=0.005;R=0.293,P=0.002)。湖羊羔羊7日龄补饲开食料能促进瘤胃乳头发育,瘤胃乳头生长相关基因调控其发育过程。
The purpose of this study was to investigate the effects of different feeding days on rumen morphology and expression of rumen epidermal growth-related genes in Hu sheep. This experiment using two factors experimental design, feeding day and lamb day two factors. A total of 78 lambs were selected to have a birth weight of (3.41 ± 0.27) kg and were randomly slaughtered at 0 days. According to the principle of homogeneity, 72 remaining lambs were divided into 7d feeding group (3.36 ± 0.15) kg) and 42d feeding group (3.44 ± 0.44) kg). 7d feeding group of lambs from 7 days old feeding feed materials 1,42d feeding group of lambs 42 days old fed feeding materials 1, two groups of lambs at 56 days of weaning, 60 days of age began to gradually open food material 2, Transition period 10d. Six lambs were randomly selected from each of the two groups at 14, 28, 42, 56, 70, and 84 days of age for slaughter. The rumen abdominis was rapidly collected and the development of rumen epidermal growth-related gene expression was analyzed by Real-time qPCR Changes were made and sections were prepared to observe rumen development. The results showed that the rumen papilla width of 7d diet group was significantly higher than that of 42d group (P <0.05), but the thickness of rumen muscle layer was significantly lower than that of 42d group (P <0.05). The expression of IGFBP3 in rumen epithelium of 7d supplemented group was significantly lower than that in 42d supplemented lamb group (P <0.05). The expression of IGFBP5 in rumen epithelium was significantly higher than that of 42d supplemented lamb group (P <0.05). The length and width of rumen papilla in 7d diet group were significantly and positively correlated with the expression of TGFβ1 and IGFBP3 in ruminal epithelium (R = 0.507, P <0.001; R = 0.444, P <0.001; , P = 0.011), but negatively correlated with the expression of IGFBP6 in rumen epithelium (R = -0.443, P <0.001; R = -0.447, P <0.001) (R = 0.227, P = 0.018). The ruminal papilla width was positively correlated with the expression of IGFBP3 and IGFBP5 in the rumen epithelium (R = 0.338, P = 0.005; R = 0.293, P = 0.002). 7-day-old lamb mutton feeding feed can promote the development of rumen papilla, rumen nipple growth-related genes regulate its development.
