目的检测Polo-like激酶1(Polo-like kinase 1,PLK1)在人类3种前列腺癌细胞株中的表达。方法采用RT-PCR和Western Blot方法检测人类前列腺癌细胞株LNCaP,DU145和PC3中PLK1 mRNA和蛋白的表达。结果在LNCaP,DU145和PC3中PLK1 mRNA均呈高表达,其PLK1/-actin比值分别为1.34,1.31,1.37;PLK1蛋白的表达水平有差异,Western Blot结果显示PLK1/-actin比值分别为1.17,0.83,0.76。结论mRNA水平上,PLK1在3种前列腺癌细胞中均呈高表达;蛋白水平上,PLK1的表达在不同前列腺癌细胞中存在差异。
Objective To detect the expression of Polo-like kinase 1 (PLK1) in three human prostate cancer cell lines. Methods The expressions of PLK1 mRNA and protein in human prostate cancer cell lines LNCaP, DU145 and PC3 were detected by RT-PCR and Western Blot. Results The PLK1 mRNA in LNCaP, DU145 and PC3 were all highly expressed, and the PLK1 / -actin ratios were 1.34, 1.31 and 1.37, respectively. The expression levels of PLK1 protein were different. Western Blot showed that the PLK1 / -actin ratios were 1.17, 0.83, 0.76. Conclusions PLK1 was highly expressed in all the three kinds of prostate cancer cells at the mRNA level. The protein level of PLK1 was different in different prostate cancer cells.
