目的研究环境持久性有机污染物邻苯二甲酸二(2-乙基己基)酯(Di-(2-ethylhexyl)phthalate,DEHP)对调节性T细胞(regulatory T cells,Treg)分化比例及其抑制功能的影响。方法构建生长期小鼠DEHP暴露模型,采用流式细胞术检测脾脏CD4+CD25+Foxp3+Treg细胞数量;免疫磁珠分选法制备CD4+CD25+T细胞和CD4+CD25-T细胞,CCK法检测CD4+CD25+T细胞和CD4+CD25-T细胞增殖能力,混合淋巴细胞反应检测CD4+CD25+T细胞对CD4+CD25-T细胞增殖的抑制作用。结果生长期小鼠经DEHP暴露处理后,CD4+CD25+Foxp3+Treg细胞数量逐渐减少,CD4+CD25+T细胞体外增殖能力及其抑制CD4+CD25-T细胞增殖的作用均逐渐降低。结论 DEHP能够从数量和功能两方面下调CD4+CD25+Treg细胞分化比例及其对效应性细胞的抑制功能。
Objective To investigate the effect of environmental persistent organic pollutants Di- (2-ethylhexyl) phthalate (DEHP) on the regulatory T cells (Treg) differentiation and its inhibition Effect of function. Methods The DEHP exposure model was established in the growing mice. The number of CD4 + CD25 + Foxp3 + Treg cells in the spleen was detected by flow cytometry. CD4 + CD25 + T cells and CD4 + CD25-T cells were prepared by magnetic bead sorting. The proliferation of CD4 + CD25 + T cells and CD4 + CD25-T cells were detected by mixed lymphocyte reaction. The inhibitory effects of CD4 + CD25 + T cells on the proliferation of CD4 + CD25-T cells were detected. Results After DEHP treatment, the number of CD4 + CD25 + Foxp3 + Treg cells gradually decreased. The proliferation of CD4 + CD25 + T cells and the inhibition of CD4 + CD25-T cell proliferation decreased gradually. Conclusion DEHP can down-regulate the proportion of CD4 + CD25 + Treg cells and their inhibitory effect on effector cells in both quantity and function.
