β-葡萄糖苷酶和β-樱草糖苷酶是茶叶香气释放过程中2个重要的内源性水解酶。采用实时定量PCR技术,分析了‘龙井43’茶品种春季不同嫩度新梢,以及8个不同品种春季一芽二叶的β-葡萄糖苷酶和β-樱草糖苷酶基因的表达差异。结果表明,β-葡萄糖苷酶基因表达量以一芽三叶最高,为5.83×107copy.μL-1,一芽三叶>一芽一叶>一芽二叶>一芽四叶;β-樱草糖苷酶基因在一芽一叶表达量很低,一芽二叶最高(2.49×106copy.μL-1),而后随着成熟度的增加而降低。β-葡萄糖苷酶基因的表达量明显高于β-樱草糖苷酶基因,一芽二叶和一芽三叶新梢中两个基因的表达量均较高。8个不同品种中,‘福鼎大白茶’2个基因表达量均非常高。8个品种β-葡萄糖苷酶基因表达量是β-樱草糖苷酶的20.6(2.4~45.6)倍。不过,‘龙井43’不同时空、不同茶树品种β-葡萄糖苷酶活性测定结果与基因表达量之间的相关性均不明显。
β-glucosidase and β-primulin glucosidase are two important endogenous hydrolases during tea aroma release. Using real-time quantitative PCR, we analyzed the expression differences of β-glucosidase and β-primulin glucosidase genes in spring tender varieties of Longjing 43 ’spring and eight spring leaves and one bud of two different varieties. The results showed that the expression of β-glucosidase gene was the highest in the three buds of one bud, which was 5.83 × 107copy.μL-1, one bud one leaf> one bud leaf> one bud leaf> one bud leaf; The expression of glycosidase gene was very low in one leaf, the highest in two buds (2.49 × 106copy.μL-1), and then decreased with the increase of maturity. The expression of β-glucosidase gene was significantly higher than that of β-primulin glucosidase gene, and the expression of both genes was higher in one bud and two buds. Eight different varieties, ’Fuding Da Bai Cha’ 2 gene expression levels are very high. Eight varieties of β-glucosidase gene expression β-primulin glycosidase 20.6 (2.4 ~ 45.6) times. However, there was no significant correlation between β-glucosidase activity assay and gene expression in different spatio-temporal varieties of Longjing43.
