目的:探讨乳腺术后骨转方对骨癌痛大鼠疼痛行为及脊髓星形胶质细胞神经胶质纤维酸性蛋白(GFAP)表达的影响。方法:SD大鼠54只,按体重随机分成6组,每组9只,于大鼠左后肢注射Walker-256细胞建立骨癌痛模型,观察各组大鼠造模后热痛敏阈值的变化,28 d后取大鼠脊髓腰膨大处,检测GFAP mRNA和蛋白、SP和CGRP蛋白的表达情况。结果:(1)乳腺术后骨转方各剂量组在造模后第21天的PWTL高于模型组(P<0.05),至造模后28 d,乳腺术后方高剂量+唑来膦酸组PWTL仍高于Model组(P<0.05)。(2)乳腺术后骨转方各剂量组可以抑制造模后28 d脊髓中GFAP mRNA和蛋白的表达,与Model组比较有统计学意义(P<0.05)。Z+H组和H组的SP蛋白表达明显低于Model组,统计学上差异显著(P<0.05);各给药组可以抑制脊髓中CGRP蛋白的表达(P<0.01)。其中Z+H组作用优于其他各给药组,统计学上差异显著(P<0.05)。结论:乳腺术后骨转方具有镇痛作用,抑制脊髓水平星形胶质细胞GFAP、SP、CGRP的表达可能是其作用机制之一。
Objective: To investigate the effect of GGZ on the pain behavior and glial glial fibrillary acidic protein (GFAP) expression of spinal cord in rats with bone cancer pain. Methods: Fifty-four Sprague-Dawley rats were randomly divided into 6 groups according to body weight, with 9 rats in each group. Walker-256 cells were injected into the left hind limbs of rats to establish bone cancer pain model. The change of thermal hyperalgesia threshold After 28 days, the lumbar enlargement of spinal cord of rats was taken and the expression of GFAP mRNA and protein, SP and CGRP protein were detected. Results: (1) The PWTL of each group on the 21st day after the operation of GOST were higher than that of the model group (P <0.05), and the high dose of zoledronic acid PWTL in acid group was still higher than Model group (P <0.05). (2) The expressions of GFAP mRNA and protein in the spinal cord after 28 days of operation were significantly decreased in each dose group (P <0.05). The protein expression of SP in Z + H group and H group was significantly lower than that in Model group (P <0.05). The expression of CGRP protein in spinal cord of each group was significantly decreased (P <0.01). Among them, the effect of Z + H group was better than other groups, the difference was statistically significant (P <0.05). CONCLUSION: Guzhuan Fang has the analgesic effect after mammary gland operation and inhibits the expression of GFAP, SP and CGRP in the spinal cord astrocyte may be one of its mechanisms.
