The authors propose a new hypothesis - the DNA hypothesis - that primary abnormalities in ALS motoneurons are abnormal DNA accumulations that can not be normally transcribed and translated and thus lead to aberrant metabolic functions and cell death in neurons. This abnormal DNA may originate from the lack of some repair mechanism of DNA in motor neurons.
