[目的]探讨不同病理学级别人脑星形细胞瘤中Rho-GT Pases家族相关Rac1/Cdc42/PAK1信号通路的作用。[方法]应用免疫组化法检测60例不同病理级别人脑星形细胞瘤组织中Rac1、Cdc42、PAK1蛋白表达,评估其与星形细胞瘤的病理临床特征的关系。[结果]①PAK1蛋白在瘤旁脑组织表达的阳性率为100.0%,在低级别和高级别星形细胞瘤中表达的阳性率分别为100.0%和50.0%,高表达率分别为60.0%和20.0%;高级别组的阳性率和高表达率均低于瘤旁脑组织和低级别组(P<0.05),且PAK1蛋白的高表达率有从瘤旁脑组织→低级别星形细胞瘤→高级别星形细胞瘤递减的趋势。②PAK1蛋白表达的阳性率在≤40岁组显著高于>40岁组,两者差别有统计学意义(P<0.05);PAK1蛋白表达的高表达率在≤40岁组与>40岁组中的差别无统计学意义(P>0.05);③PAK1蛋白表达的阳性率和高表达率与性别、部位及肿瘤最大径无关(P>0.05);④Rac1、Cdc42蛋白均为阴性表达。[结论]PAK1在星形细胞瘤中的作用可能与Rac1/Cdc42/PAK1信号通路关系不大,而可能是通过PAK1相关其他通路起作用;PAK1在了解星形细胞瘤的发生、进展和寻找治疗新靶点方面可能有参考价值。
[Objective] To investigate the role of Rac1 / Cdc42 / PAK1 signaling pathway in Rho-GT Pases family in human astrocytomas of different pathological grades. [Method] The expressions of Rac1, Cdc42 and PAK1 protein in 60 cases of human astrocytomas with different pathological grades were detected by immunohistochemistry, and their relationship with pathological features of astrocytomas was evaluated. [Results] ① The positive rate of PAK1 protein expression in tumor tissues was 100.0%. The positive rates of expression of PAK1 protein in low grade and high grade astrocytomas were 100.0% and 50.0%, respectively, and the high expression rates were 60.0% and 20.0% respectively %. The positive rate and high expression rate of high grade group were lower than that of the tumor-adjacent brain tissue and the low grade group (P <0.05), and the high expression rate of PAK1 protein from the tumor tissue → low grade astrocytoma → High-grade astrocytoma decreasing trend. (2) The positive rate of PAK1 expression in patients ≤40 years old was significantly higher than> 40 years old, the difference was statistically significant (P <0.05); The high expression rate of PAK1 protein was less than 40 years old and> 40 years old (P> 0.05). ③ The positive rate and high expression rate of PAK1 protein were not related to the sex, location and tumor diameter (P> 0.05). ④ The expressions of Rac1 and Cdc42 were all negative. [Conclusion] The role of PAK1 in astrocytoma may not be related to the Rac1 / Cdc42 / PAK1 signaling pathway, but may play a role in other pathways related to PAK1. PAK1 may play an important role in understanding the occurrence, progression and seeking treatment of astrocytoma New target may have reference value.
