目的:建立HPLC法测定经过砷胁迫处理栽培的黄芩根中5个黄酮类成分的含量。方法:采用C18色谱柱,流动相为乙腈-0.1%甲酸水溶液,检测波长为275nm,流速1.0mL·min-1,柱温27℃。结果:5个黄酮类成分在30min内可达到完全分离,每种成分在各自的检测浓度范围内线性关系良好。结论:本法操作简便,结果准确,可以同时测定黄芩中5个黄酮类成分的含量,而且首次应用于砷胁迫下黄芩中药效成分的量化分析,适用于黄芩类药材的成分分析及质量控制。
Objective: To establish a HPLC method for the determination of five flavonoids in Scutellaria baicalensis roots treated with arsenic stress. Methods: A C18 column was used. The mobile phase was acetonitrile-0.1% formic acid aqueous solution. The detection wavelength was 275 nm, the flow rate was 1.0 mL·min-1, and the column temperature was 27°C. RESULTS: Five flavonoids could be completely separated within 30 min, and each component had a good linear relationship within their respective detection concentration range. Conclusion: This method is simple, accurate, and can simultaneously determine the content of five flavonoids in Astragalus membranaceus. It is also the first to be applied to the quantitative analysis of the effective components of Astragalus membranaceus under arsenic stress. It is suitable for composition analysis and quality control of Astragalus membranaceus. .
