目的探讨醒脑静合生脉注射液对大鼠脑出血后脑组织内凝血酶受体-1(PAR1)表达的影响。方法将40只雄性SD大鼠随机分为脑出血组(ICH)、生理盐水组(NS)、醒脑静合生脉注射液治疗组(XNJSM)、水蛭素治疗组(HIR),每组10只大鼠。采用自体不凝血注入法建立脑出血模型。术后72h取脑组织,HE染色观察各组血肿周围神经细胞形态学改变,免疫组织化学方法及Western blotting法检测各组血肿周围脑组织PAR1的表达。结果脑出血后血肿周围脑组织PAR1表达增强,XNJSM组、HIR组脑组织病理形态明显改善,脑组织PAR1表达减少,与ICH组比较差异有显著性(P<0.05)。结论醒脑静合生脉注射液可能通过凝血酶受体-1途径有效抑制大鼠脑出血后PAR1蛋白表达,对脑出血后脑组织发挥保护作用。
Objective To investigate the effect of xingnaojing shenmai injection on the expression of thrombin receptor 1 (PAR1) in brain tissue after intracerebral hemorrhage in rats. Methods Forty male SD rats were randomly divided into intracerebral hemorrhage (ICH) group, NS group, XNJSM group, and hirudin group (n = 10) Rat. Intracerebral hemorrhage model was established by self-non-coagulation infusion. The brain tissue was harvested at 72 hours after operation. The morphological changes of the peripheral nerve cells in each group were observed by HE staining. The expression of PAR1 in the perihematomal brain tissue was detected by immunohistochemistry and Western blotting. Results Cerebral hemorrhage increased the expression of PAR1 in brain tissue around the hematoma. The pathological changes of brain tissue in XNJSM group and HIR group were significantly improved. The expression of PAR1 in brain tissue decreased compared with ICH group (P <0.05). Conclusion Xingnao Jinghe Shengmai Injection can effectively inhibit the expression of PAR1 protein after intracerebral hemorrhage through the thrombin receptor - 1 pathway and play a protective role in the brain tissue after intracerebral hemorrhage.
