目的初步研究体外5-氨基酮戊酸介导的光动力疗法对人舌鳞癌Tca8113细胞的杀伤效应。方法体外培养Tca8113细胞,以5-氨基酮戊酸为光敏剂,半导体激光治疗仪给予光动力疗法,采用MTT法检测光敏剂不同孵育时间、不同光敏剂浓度对Tca8113细胞抑制率的影响。结果 5-ALA-PDT作用后,Tca8113细胞生长受到抑制,孵育时间和浓度效应关系显著(P<0.05)。药物最佳作用浓度为1mmol/l,最佳孵育时间为6h。结论 5-氨基酮戊酸-光动力疗法能有效杀伤Tca8113细胞,光敏剂孵育时间、光敏剂浓度是影响疗效的重要因素。
Objective To study the killing effect of 5-aminolevulinic acid-mediated photodynamic therapy on Tca8113 human tongue squamous cell carcinoma in vitro. Methods Tca8113 cells were cultured in vitro. 5-Aminolevulinic acid was used as photosensitizer and photodynamic therapy was performed by semiconductor laser treatment. The effects of different photosensitizer incubation time and different photosensitizer concentrations on the inhibition rate of Tca8113 cells were detected by MTT assay. Results After 5-ALA-PDT treatment, the growth of Tca8113 cells was inhibited. The incubation time and concentration effect were significantly correlated (P <0.05). The optimal concentration of the drug was 1mmol / l, the best incubation time was 6h. Conclusion 5-aminolevulinic acid-photodynamic therapy can effectively kill Tca8113 cells, photosensitizer incubation time, photosensitizer concentration is an important factor affecting the curative effect.
