目的: 探讨姜黄素对急性早幼粒细胞白血病细胞株NB4细胞的增殖抑制作用及其作用机制。方法:以 0, 5, 10, 20, 30μmol·L-1浓度的姜黄素作用于体外培养的NB4细胞,分别于培养后 12, 24,36, 48, 60, 72h用四氮唑蓝比色法检测细胞生长抑制率,应用流式细胞仪及Hoechst33258荧光染色法观察细胞凋亡,采用聚合酶链反应 酶标记免疫吸附法检测细胞凋亡前后的端粒酶活性。结果: 10 ~30mol·L-1浓度的姜黄素作用 24 ~72h可显著降低NB4细胞端粒酶活性,抑制细胞的生长,诱导细胞发生凋亡。姜黄素对细胞的增殖抑制作用呈现出一定的剂量 效应与时间 效应关系, 细胞凋亡率在药物作用 60h左右达高峰。结论:姜黄素具有显著的体外抗白血病作用,降低细胞端粒酶活性可能是其重要作用机制之一。
Objective: To investigate the inhibitory effect of curcumin on the proliferation of acute promyelocytic leukemia NB4 cells and its mechanism. Methods: Curcumin at concentrations of 0, 5, 10, 20 and 30 μmol·L -1 were used to in vitro cultured NB4 cells. The cells were stained with tetrazolium blue at 12, 24, 36, 48, 60, The cell growth inhibition rate was detected by flow cytometry and Hoechst33258 fluorescence staining. The telomerase activity before and after apoptosis was detected by polymerase chain reaction and enzyme-linked immunosorbent assay (ELISA). Results: The effect of curcumin at concentration of 10 ~ 30mol·L-1 for 24h ~ 72h could significantly decrease the telomerase activity, inhibit the growth of NB4 cells and induce the apoptosis of NB4 cells. The inhibitory effect of curcumin on cell proliferation showed a certain dose-effect and time-dependent effects, the rate of apoptosis in the drug reached a peak about 60h. CONCLUSION: Curcumin has significant anti-leukemia effect in vitro. It may be one of the important mechanisms of curcumin to reduce telomerase activity.
