目的:建立金益排石颗粒中大黄酚含量的微乳液相色谱测定法。方法:以微乳2.5%(w/v)十二烷基硫酸钠(SDS)-0.1%(v/v)正辛烷-10.0%(v/v)正丁醇-0.5%三乙胺(v/v)(磷酸调pH至3.0)为流动相,色谱柱采用Hypersil ODS_2色谱柱(250 mm×4.6 mm,5μm),柱温28℃,流速1 ml·min~(-1),检测波长254 nm。结果:大黄酚在3.9~39.0μg·ml~(-1)范围内,与峰面积呈良好的线性关系(r=0.999 8),平均加样回收率为99.66%,RSD为0.64%(n=6)。结论:本法简便、准确,可用于金益排石颗粒中大黄酚的含量测定。
OBJECTIVE: To establish a microemulsion liquid chromatographic method for determination of chrysophanol in Jinyi Paishi granules. Methods: The microemulsion was prepared with 2.5% (w / v) sodium dodecyl sulfate (SDS) - 0.1% (v / v) n-octane -10.0% (v / v) n-butanol -0.5% v / v) (adjusted to pH 3.0 with phosphoric acid) as mobile phase. The column was Hypersil ODS_2 (250 mm × 4.6 mm, 5 μm) with a column temperature of 28 ℃ and a flow rate of 1 ml · min ~ 254 nm. Results: In the range of 3.9 ~ 39.0μg · ml ~ (-1), chymol had a good linear relationship with the peak area (r = 0.999 8), the average recovery was 99.66% and the RSD was 0.64% (n = 6). Conclusion: This method is simple, accurate, and can be used for the determination of chrysophanol in Jinyi Paishi granules.
