用伴刀豆凝集素(ConA)-辣根过氧化物酶(HRP)定位技术观察乌拉坦经胎盘对胚胎鼠及新生鼠肝细胞ConA受体分布变化的影响。结果显示:(1)13~15 d胚胎肝细胞ConA受体主要分布于细胞膜及核膜;17~19 d胚胎肝细胞胞膜ConA受体增多,核膜受体消失,胞浆出现ConA受体;3~14 d新生鼠ConA受体主要分布于胞浆,胞膜和核膜的ConA受体基本消失。(2)用乌拉坦后可减慢胞膜上ConA受体的消失,延迟胞浆ConA受体的出现,尤其对11~15 d胚胎作用最为明显。提示正常胚胎肝细胞ConA受体分布随肝细胞成熟呈规律性变化;乌拉坦可能在分子水平干扰了胚胎肝细胞的正常发育与分化。
The effects of urethane on the distribution of ConA receptor in embryonic and neonatal rat hepatocytes were observed by ConA - HRP localization technique. The results showed that: (1) ConA receptors of embryonic hepatocytes were mainly distributed on the cell membrane and nuclear membrane from 13 to 15 days; on day 17 to 19, the number of ConA receptors on the membrane of hepatocytes was increased, the nuclear membrane receptors disappeared, and ConA receptors ConA receptors in neonatal rats were mainly distributed in the cytoplasm from 3 to 14 days, and ConA receptors in the plasma membrane and nuclear membrane disappeared. (2) The use of urethane slowed down the disappearance of ConA receptors on the membrane, delayed the appearance of the ConA receptors in cytoplasm, especially on embryos from 11 to 15 days. Suggesting that the distribution of ConA receptors in normal embryonic hepatocytes changes regularly with the maturation of hepatocytes. Ulatan may interfere with the normal development and differentiation of embryonic hepatocytes at the molecular level.
