目的探讨黄芪注射液对肝癌细胞生长及细胞内NF-κB蛋白表达的影响。方法采用系列浓度的黄芪注射液作用HepG2细胞,以MTT法检测黄芪注射液对HepG2细胞生长的影响。根据MTT实验结果,分别采用浓度为0,6.25%,12.5%,25.0%的黄芪注射液作用HepG2细胞48 h,West-ern blot法检测各组肝癌细胞中NF-κB的表达。结果不同浓度黄芪注射液对HepG2细胞的生长均有抑制作用,并呈浓度依赖性(P<0.05~0.01),其半数抑制浓度(IC50)为0.39192。Western blot检测显示,黄芪注射液能浓度依赖性抑制HepG2细胞中NF-κB蛋白表达。结论黄芪注射液对肝癌HepG2细胞的生长有抑制作用,并能下调细胞NF-κB蛋白的表达。
Objective To investigate the effect of Astragalus injection on the growth of hepatocellular carcinoma cells and the expression of NF-κB protein. Methods HepG2 cells were treated with a series of concentrations of astragalus injection and the effect of astragalus injection on the growth of HepG2 cells was detected by MTT assay. According to MTT results, HepG2 cells were treated with Astragalus membranaceus at concentrations of 0, 6.25%, 12.5% and 25.0% respectively for 48 h. The expression of NF-κB in each group was detected by West-ern blot. Results Astragalus membranaceus injection with different concentrations inhibited the growth of HepG2 cells in a concentration - dependent manner (P <0.05 ~ 0.01). The IC50 was 0.39192. Western blot showed that Astragalus injection could inhibit the expression of NF-κB in HepG2 cells in a concentration-dependent manner. Conclusion Astragalus injection can inhibit the growth of HepG2 cells and down-regulate the expression of NF-κB protein.
