心脉龙干预后氧自由基代谢及脑、肝组织脂褐质含量的变化(英文)

背景:心脉龙是从昆虫类中药美洲大蠊中发现并提纯制备的新型核苷类化合物。通过大量的实验研究表明,具有明显的心血管活性,可用于治疗急慢性心力衰竭及休克,明显改善微循环。目的:观察纯中药制剂心脉龙的抗衰老作用。设计:对照观察实验。单位:大理学院基础医学院生理学与病理生理学教研室。材料:实验于2002-07/2004-09在大理学院基础医学院机能学实验室完成。选用ICR小鼠40只,雌雄不拘,由昆明医学院云南省天然药物药理重点实验室提供。心脉龙注射液由云南腾冲制药厂生产[批号:94(ZL)03]。方法:40只ICR小鼠采用抽签法随机分为4组:青年对照组,衰老对照组,心脉龙大剂量组和心脉龙小剂量组,每组10只。青年对照组为3月龄小鼠,体质量(20±2)g,其余3组均饲养20月龄,体质量(40±3)g。从第20月开始,心脉龙小、大剂量组分别按6mg/kg、8mg/kg连续腹腔注射心脉龙30d,1次/d。青年和衰老对照组腹腔注射相同剂量的生理盐水。第31天,将实验小鼠断头取血、脑和肝脏,制成10%组织匀浆。按试剂盒说明书中的方法测定血清过氧化氢酶、超氧化物歧化酶和谷胱甘肽过氧化物酶活性,测定脑组织丙二醛含量及脑组织和肝脏脂褐质含量。主要观察指标:①血清过氧化氢酶、超氧化物歧化酶和谷胱甘肽过氧化物酶活性。②脑组织丙二醛含量及脑组织和肝脏脂褐质含量。结果:40只实验动物均进入结果分析,无脱失。①衰老对照组小鼠血清过氧化氢酶、超氧化物歧化酶和谷胱甘肽过氧化物酶活性明显低于青年对照组,而心脉龙两剂量组均能显著提高老年小鼠血清过氧化氢酶、超氧化物歧化酶和谷胱甘肽过氧化物酶活性,与衰老对照组比较,差异有显著性意义(P<0.05或P<0.01)。②衰老对照组小鼠脑组织丙二醛含量及脑组织和肝脏脂褐质含量明显高于青年对照组,而心脉龙大、小剂量组均能明显降低脑组织丙二醛含量及脑组织和肝脏脂褐质含量,与衰老对照组比较差异有非常显著性意义(P<0.01)。但心脉龙两剂量组差异无显著性意义。结论:心脉龙可通过改善氧自由基代谢而发挥抗衰老作用。 Background: Xinmailong is a novel nucleoside compound that has been found and purified from insect cockroach American cockroach. Through a large number of experimental studies have shown that there is significant cardiovascular activity, can be used to treat acute and chronic heart failure and shock, significantly improve the microcirculation. Objective: To observe the anti-aging effect of pure Chinese medicine preparation Xinmailong. Design: Control observation experiment. Unit: Department of Physiology and Pathophysiology, Basic Medical College, Dali University. MATERIALS: The experiment was performed at the Functional Laboratory of the Basic Medical College of Dali University from July 2002 to September 2004. 40 ICR mice were selected, either male or female, and provided by Yunnan Provincial Key Laboratory of Pharmacology of Natural Medicines, Kunming Medical College. Xinmailong injection was produced by Yunnan Tengchong Pharmaceutical Factory [lot: 94(ZL)03]. Methods: Forty ICR mice were randomly divided into 4 groups: the young control group, the aging control group, the high-dose Xinmailong group and the low-dose Xinmailong group, with 10 rats in each group. The young control group was a 3-month-old mouse, with a body weight of (20±2) g. The remaining three groups were kept for 20 months and the body weight was (40±3) g. From the 20th month, Xinmai Longxiao and large dose groups were continuously injected intraperitoneally with 6mg/kg and 8mg/kg respectively for 30 days, once a day. Youth and senile control group were injected intraperitoneally with the same dose of physiological saline. On the 31st day, the experimental mice were decapitated to take blood, brain and liver to make a 10% tissue homogenate. Serum catalase, superoxide dismutase and glutathione peroxidase activities were determined according to the method described in the kit instructions. Malondialdehyde content in brain tissue and lipofuscin content in brain tissue and liver were determined. MAIN OUTCOME MEASURES: Serum catalase, superoxide dismutase and glutathione peroxidase activity. 2 Malondialdehyde content in brain tissue and lipofuscin content in brain tissue and liver. RESULTS: Forty experimental animals all entered the results analysis without loss. 1 The activities of serum catalase, superoxide dismutase and glutathione peroxidase in the senescent control group were significantly lower than those in the young control group, but the two groups of Xinmailong could significantly increase the serum of old mice. Oxygenase, superoxide dismutase and glutathione peroxidase activities were significantly different from those in the senescent control group (P<0.05 or P<0.01). 2 The content of malondialdehyde in the brain and the content of lipofuscin in the brain and liver of the senile control group mice were significantly higher than that of the young control group. Both the large and small doses of Xinmai Longda significantly reduced the content of malondialdehyde in the brain and brain tissue. The lipofuscin content in the liver and the senescent control group were significantly different (P<0.01). However, there was no significant difference in the two groups of Xinmailong. Conclusion: Xinmailong can play an anti-aging effect by improving the metabolism of oxygen free radicals.
