目的:建立反相高效液相色谱法测定新疆蓝刺头花乙醇提取物中芦丁含量的方法。方法:色谱柱(C18,250mm×4.6mm,5μm),流动相为甲醇-0.4%磷酸(50∶50),流速0.6mL/min,柱温25℃,检测波长360nm,进样量5μL。结果:芦丁在0.005~0.03μg范围内线性良好(r=0.9997),平均回收率为99.0%,RSD为1.25%(n=5)。结论:文中所建方法简单、快速、精密度高、重复性好,对新疆蓝刺头资源的开发利用具有一定的价值,对此种民族药材的质量控制研究提供参考。
Objective: To establish a method for the determination of rutin content in ethanol extract of blue spinhead flower in Xinjiang by reversed-phase high performance liquid chromatography. METHODS: The mobile phase was methanol-0.4% phosphoric acid (50:50) at a flow rate of 0.6 mL / min and the column temperature was 25 ℃. The detection wavelength was 360 nm and the injection volume was 5 μL. RESULTS: Rutin showed good linearity (r = 0.9997) in the range of 0.005-0.03 μg with an average recovery of 99.0% and a RSD of 1.25% (n = 5). Conclusion: The method proposed in this paper is simple, rapid, accurate and reproducible. It is of great value to the development and utilization of blue prickly resources in Xinjiang and provides reference for the quality control of this species.
