目的探讨口腔种植体周围炎对颌骨成骨细胞增殖分化能力的影响,为临床治疗提供参考。方法收集医院口腔科松动并拔出种植体患者7例作为观察组,拔出第三磨牙身体健康者7例作为对照组,取两组患者的颌骨组织进行成骨细胞的分离培养和纯化,MMT测定成骨细胞增殖能力,RT-PCR法测定成骨细胞Ⅰ型胶原(Ⅰcollagen,Col I)mRNA水平,Western blot检测成骨细胞Col I蛋白水平。结果第7天时,观察组成骨细胞OD值低于对照组(P<0.05),第3天时,观察组成骨细胞OD值低于对照组,但差异无统计学意义;观察组成骨细胞Col I蛋白相对表达量明显低于对照组(P<0.05)。结论口腔种植体周围炎能够抑制颌骨成骨细胞的增殖和分化。
Objective To investigate the effect of oral peri-implantitis on the proliferation and differentiation of osteoblasts in the mandible and to provide a reference for clinical treatment. Methods 7 cases of patients with loose stomatology and pulling out implants in the hospital were collected as observation group and 7 healthy people with third molars as control group.The jaw tissues of two groups were used to separate and culture osteoblasts, The proliferation of osteoblasts was measured by MMT, the mRNA level of type Ⅰ collagen (Ⅰcollagen, Col I) was detected by RT-PCR and the level of Col I protein by Western blot. Results At the 7th day, the OD value of the osteoblasts in the observation group was lower than that of the control group (P <0.05). On the third day, the OD value of the osteoblasts in the observation group was lower than that of the control group, but the difference was not statistically significant. The relative expression level was significantly lower than the control group (P <0.05). Conclusion Peri-implantitis can inhibit the proliferation and differentiation of osteoblasts in the oral cavity.
