本文的部份研究涉及用~1H-NMR 方法探讨药物的代谢作用,为了简化谱图以利解析,对处理样品用的固相提取技术的使用进行了研究。选用萘普生(Naproxen)为模型化合物。从一可疑关节炎患者口服500mg 萘普生后的女性病人,收集尿样。先进行冷冻干燥,然后溶在重水中。~1H-NMR谱于室温(25±1℃)下测定。扫描宽度为3401 Hz(在250MHz NMR仪上),数据点16384;取样时间2.4s;脉冲宽度90°;脉冲延迟5 s。为了压制残余水峰,在水的共振频率处(4.8 ppm)加上第二照射场。为了减少去偶仪噪声,在取样期间关闭去偶仪门。
Some of the studies in this paper involve the use of ~ 1H-NMR methods to investigate the metabolic effects of drugs. In order to simplify the analysis of spectra, solid-phase extraction techniques for the treatment of samples were studied. Naproxen was chosen as the model compound. Urine samples were collected from female patients after oral administration of 500 mg of naproxen to a suspected arthritis patient. Freeze-dried and then dissolved in heavy water. ~ 1H-NMR spectra were measured at room temperature (25 ± 1 ° C). The scan width was 3401 Hz (on a 250 MHz NMR instrument), data point 16384; sampling time 2.4 s; pulse width 90 °; pulse delay 5 s. In order to suppress the residual water peak, a second irradiation field is added at the resonant frequency of the water (4.8 ppm). In order to reduce the missed instrument noise, turn off the instrument gate during sampling.
