目的:探讨大肠腺癌突变型p53基因蛋白表达与p53基因cDNA突变间的相互关系和意义。方法:对100例新鲜大肠腺癌组织采用PAb240单克隆抗体免疫组织化学染色(LSAB法)检测突变型p53基因蛋白表达、RT-PCR-SSCP检测p53基因cDNA突变,并比较它们间相互关系。结果:100例大肠腺癌中,76例PAb240单抗阳性(76%),51例p53基因cDNA突变阳性(51%);PAb240单抗反应与p53基因cDNA突变比较,两者皆阳性49%,两者中一者阳性29%,两者皆阴性22%(P<0.0001)。结论:p53基因cDNA突变与其基因蛋白产物结构改变高度吻合,大肠腺癌p53基因mRNA(cDNA)突变是参与和影响突变型p53基因蛋白结构改变和生物学行为的主要因素
Objective: To investigate the relationship between the expression of mutant p53 gene protein and p53 gene cDNA mutation in colorectal adenocarcinoma. METHODS: 100 cases of fresh colorectal adenocarcinoma were examined for the expression of mutant p53 gene by the LSAB monoclonal antibody and the p53 gene mutation was detected by RT-PCR-SSCP. Their correlations were also compared. RESULTS: Of the 100 cases of colorectal adenocarcinoma, 76 cases were positive for PAb240 (76%), and 51 cases of p53 gene mutation were positive (51%). The PAb240 mAb reaction was compared with the p53 gene mutation, both of which were 49% positive. One of them was positive in 29% and both were negative in 22% (P<0.0001). Conclusion: The mutation of p53 gene is highly consistent with the structural change of its gene protein product. The p53 gene mRNA (cDNA) mutation in colorectal adenocarcinoma is the main factor involved in and affecting the structural changes and biological behavior of the mutant p53 gene protein.
