目的探讨老龄大鼠基底前脑-皮质胆碱能系统TRKB蛋白表达的老龄性变化,同时对比观察人参皂甙对其改变的影响。方法选用WISTAR♀大鼠30只,随机分为3组(N=10):青年组、老龄组、给药组(第17个月始饲以人参皂甙至25-27月龄)。采用WESTERN BLOT技术结合图像分析对基底前脑、海马结构和大脑皮质进行定量研究。结果老龄组基底前脑、海马结构和大脑皮质TRKB蛋白表达分别较青年组下调了61%、99.7%和57.7% (P<0.01),但给药组上述3个脑区的TRKB蛋白表达较老龄组有所回升,分别较老龄组上调了65%、78.5%和 22.6%(P<0.01)。结论老龄组基底前脑-皮质胆碱能系统TRKB蛋白表达较青年组下调,而人参皂甙可明显上调该3个脑区TRKB蛋白的表达。
Objective To investigate the age-related changes of TRKB protein expression in the basal forebrain-cortical cholinergic system in aged rats, and to observe the effects of ginsenosides on their changes. Methods Thirty Wistar rats were randomly divided into 3 groups (N=10): young group, old-age group, and administration group (17-month feeding of ginsenosides to 25-27 months of age). The quantitative analysis of basal forebrain, hippocampal formation and cerebral cortex was performed using Western BLOT technique combined with image analysis. Results The expression of TRKB protein in the basal forebrain, hippocampus and cerebral cortex of the aged group was lower than that of the young group by 61%, 99.7% and 57.7%, respectively (P<0.01). The TRKB protein expression in the elderly group increased compared with the old age group, which was 65%, 78.5%, and 22.6%, respectively, compared with the old group (P<0.01). CONCLUSION: The expression of TRKB protein in the basal forebrain-cortical cholinergic system in the aged group was lower than that in the young group, while ginsenosides could significantly up-regulate the expression of TRKB protein in these three brain regions.
