为探讨一氧化氮(NO)的前体L-精氨酸(L-Arg)和NO合成酶抑制剂L-硝基-精氨酸甲酯(L-NAME)对围产期窒息新生大鼠血浆内皮素(ET)的影响,采用放射免疫分析法检测窒息新生大鼠及窒息前应有L-Arg或L-NAME预处理的新生鼠血浆ET的水平。结果显示:窒息组血浆ET水平较对照组明显升高(P<0.01);L-Arg组血浆ET水平较窒息组明显下降(P<0.01),与对照组相比无显著差异(P>0.05);而L-NAME组血浆ET水平明显高于对照组及L-Arg组(P<0.01),与窒息组比较,无统计学意义。提示:L-Arg能有效地降低窒息新生大鼠血浆ET的水平;L—NAME则作用相反,NO合成抑制可明显增加ET水平。
To investigate the effects of L-arginine (L-Arg), a precursor of nitric oxide (NO), and L-NAME, an NO synthase inhibitor, on perinatal asphyxia neonatal rats Plasma endothelin (ET) was measured by radioimmunoassay in neonatal rats with asphyxia and neonatal rats pretreated with either L-Arg or L-NAME. The levels of plasma ET in asphyxia group were significantly higher than those in control group (P <0.01). The levels of plasma ET in L-Arg group were significantly lower than those in asphyxia group (P <0.01), but no significant difference compared with control group (P> 0.05). However, the plasma ET level in L-NAME group was significantly higher than that in control group and L-Arg group (P <0.01), but not in asphyxia group. It is suggested that L-Arg can effectively reduce the level of plasma ET in neonatal rats with asphyxia; while L-NAME has the opposite effect, inhibition of NO synthesis can significantly increase the level of ET.
