目的考察苦杏仁苷作为液相色谱电喷雾电离质谱联用技术测定血清中多种成分含量的内标物使用的可行性。方法血清样品加入苦杏仁苷,经三氯乙酸沉淀蛋白,以含甲酸的乙腈-水为流动相梯度洗脱,用Waters Acquity HSS T3色谱柱分离,电喷雾电离,正、负离子模式分别检测。结果在正离子和负离子模式下,血清样品中苦杏仁苷本身的峰面积与浓度线性关系良好,苦杏仁苷作为内标物测定血清中的芍药苷和阿魏酸,待测物内标峰面积比值与待测物浓度线性关系良好。结论苦杏仁苷可以作为液相色谱电喷雾电离质谱联用技术测定药后血清中多种成分含量的内标物使用。
Objective To investigate the feasibility of using amygdalin as an internal standard for determining the content of various components in serum by using liquid chromatography coupled with electrospray ionization mass spectrometry. Methods Serum samples were extracted with amygdalin, precipitated with trichloroacetic acid and eluted with formic acid in acetonitrile - water. The mobile phase was separated on a Waters Acquity HSS T3 column and electrospray ionization was performed. Results In positive and negative ion mode, the peak area and the concentration of amygdalin in serum samples were good linearity. The content of paeoniflorin and ferulic acid in serum was determined by amygdalin as internal standard, and the peak area The ratio and the analyte concentration linear relationship is good. Conclusion Amygdalin can be used as an internal standard for the determination of the content of various components in the serum of a drug by liquid chromatography-electrospray ionization mass spectrometry.
