目的:探讨藤茶双氢杨梅素对动脉粥样硬化(AS)大鼠血清炎症因子水平和ⅡA分泌型血小板型磷脂酶A2(secretory phospholipase A2of groupⅡA,s PLA2-ⅡA)mRNA表达的影响,以了解藤茶双氢杨梅素对AS的作用机制。方法:取健康雄性Wistar大鼠60只,随机分为5组,即正常组、模型组、辛伐他汀组(5 mg·kg-1·d-1)、及藤茶双氢杨梅素低、高剂量组(10,40 mg·kg-1·d-1),每组12只,除正常组大鼠喂以基础饲料外,其他组给大鼠饲养高脂饲料,并ip维生素D3,复制AS模型,给药组ig给予相应药物,正常组与模型组给予等体积的生理盐水,连续24周后,观察藤茶双氢杨梅素对模型大鼠血清C-反应蛋白(CRP),血清肿瘤坏死因子α(TNF-α)和白细胞介素-1(IL-1)的水平,RT-PCR观察主动脉弓s PLA2-ⅡA mRNA表达的变化。结果:与正常组比较,模型组大鼠血管管壁增厚,内皮粗糙,中膜平滑肌纤维明显增生,大鼠血清CRP,TNF-α和IL-1水平明显增高(P<0.05,P<0.01),主动脉弓s PLA2-ⅡA mRNA的表达明显增强(P<0.01);与模型组比较,藤茶双氢杨梅素低剂量组大鼠血管病变范围及内膜增厚程度略有减轻,高剂量组大鼠血管仅见局部内皮粗糙,内膜光滑,中膜平滑肌纤维轻度增生,藤茶双氢杨梅素高剂量组可明显降低AS大鼠血清CRP和IL-1水平(P<0.05),能显著降低AS大鼠主动脉弓s PLA2-ⅡA mRNA的表达(P<0.05)。结论:藤茶双氢杨梅素对AS具有一定的防治作用,其作用机制是通过改善AS大鼠炎症因子水平,抑制s PLA2-ⅡA mRNA的表达。

Results: Compared with the normal group, the vascular wall thickening, rough endothelium and smooth muscle fibers in the model group were significantly increased, and the levels of serum CRP, TNF-α and IL-1 in the model group were significantly increased (P <0.05, P <0.01) ), PLA2-ⅡA mRNA expression in aortic arch significantly increased (P <0.01) .Compared with the model group, the vasoconstriction and intimal thickening of rats in the low-dose ramie group were slightly reduced, while the high dose group Rat blood vessels only local rough, smooth intima, smooth muscle fibers mild hyperplasia, Rattan high doses of dihydromyricetin can significantly reduce serum levels of CRP and IL-1 (P <0.05), can be significant Reduce the expression of PLA2-ⅡA mRNA in AS aortic arch (P <0.05). CONCLUSION: Rattan tea dihydromyricetin has a preventive and therapeutic effect on AS, and its mechanism is to inhibit the expression of PLA2-ⅡA mRNA by improving the level of inflammatory cytokines in rats.
