目的:观察黄芪注射液对兔脂肪来源的间充质干细胞(ASCs)体外增殖的作用。方法:提取家兔脂肪间充质干细胞,培养后,设实验组和对照组,实验组加入不同质量浓度黄芪注射液,对照组加常规培养液,用流式细胞仪观察细胞周期,用四唑盐(MTT)法测定细胞的增殖率,比较两组的细胞周期和细胞增殖率。结果:培养24 h后,对照组增殖期的细胞占(65.3±4.8)%,黄芪注射液组占(72.6±5.7)%,差异有统计学意义(P<0.05);培养48 h后,黄芪注射液0.390 6~200 g.L-1,细胞增殖(吸光度A)为(2.273±0.100)~(1.983±0.125),对照组为(1.341±0.424),表明实验范围内各浓度黄芪注射液对ASCs均有明显促进增殖的作用,两组比较差异有统计学意义(P<0.01)。结论:黄芪注射液有明显的促进兔ASCs体外增殖的作用。
Objective: To observe the effect of Astragalus injection on proliferation of adipose derived mesenchymal stem cells (ASCs) in vitro. Methods: Adipose-derived mesenchymal stem cells were isolated from rabbits and cultured. The experimental group and the control group were set up. The experimental groups were given Huangqi injection of different mass concentration. The control group was given routine culture medium. The cell cycle was observed by flow cytometry. The cell proliferation rate was measured by MTT method. The cell cycle and cell proliferation rate were compared between the two groups. Results: After cultured for 24 h, cells in the control group proliferated (65.3 ± 4.8)% and Astragalus injection group (72.6 ± 5.7)%, the difference was statistically significant (P <0.05) (2.273 ± 0.100) ~ (1.983 ± 0.125) in control group and (1.341 ± 0.424) in control group with 0.390 6 ~ 200 gL-1 injection, indicating that the concentration of Astragalus membranaceus in each experimental group There was a significant difference between the two groups (P <0.01). Conclusion: Astragalus injection can significantly promote the proliferation of rabbit ASCs in vitro.
