本研究以芥菜型油菜(Brassica juncea L.)核不育系1161A和温敏雄性不育系K121S的BCF1为作图群体,采用混合群体分组分析法(bulked segregant analysis,BSA)定位油菜温敏雄性不育系K121S的育性转换基因(Fc)。研究结果表明,从86对多态性SSR引物中获得了5个连锁标记:Ol11-G11a、Na10-F06、Na14-G10、CN48和Br GMS961,其中Ol11-G11a、Na14-G10和Na10-F06等3个SSR标记与K121S育性转换基因的遗传距离较近,分别为1.6 c M、11.0 c M和19.7 c M。本研究结果可为油菜温敏雄性不育系K121S的育性转换基因的图位克隆及其利用提供依据。
In this study, Brassica juncea L. male sterile line 1161A and thermo-sensitive male sterile line K121S BCF1 were used as a mapping population, and a mixed population grouping analysis (BSA) was used to map the temperature-sensitive male Fertility Transform Gene (Fc) in CMS Line K121S. The results showed that five linkage markers were obtained from 86 polymorphic SSR primers: Ol11-G11a, Na10-F06, Na14-G10, CN48 and Br GMS961, among which Ol11-G11a, Na14-G10 and Na10-F06 The genetic distances of the three SSR markers to the K121S fertility transgene were 1.6 cM, 11.0 cM and 19.7 cM, respectively. The results of this study may provide the basis for the mapping and utilization of the fertility-switching genes of the thermo-sensitive male sterile line K121S in rapeseed.
