目的揭示巨噬细胞集落刺激因子 (M- CSF)是否能保护单核巨噬细胞免受过氧化损伤。方法以 L 92 9细胞条件培养液 (L92 9- CM)作为 M- CSF来源 ,以细胞显微形态、细胞活力等指标 ,观察了 M- CSF对叔丁基氢过氧化物 (tb OOH)诱导的巨噬细胞系 RAW2 6 4.7细胞氧化损伤的影响。结果 tb OOH可以引起 RAW2 6 4.7细胞的损伤 ,表现为细胞皱缩、伪足消失、细胞活力降低 ,且损伤随 tb OOH作用浓度的增加而明显 ;而 M- CSF可以减轻 tb OOH诱发的 RAW2 6 4.7细胞氧化损伤。结论 M-CSF对单核巨噬细胞的氧化损伤具有保护作用。
Objective To reveal whether macrophage colony stimulating factor (M-CSF) protects monocytes from peroxisome damage. Methods L 92 9-CM was used as the source of M-CSF in L 92 9 cell culture medium. The effects of M-CSF on tb OOH induced macrophages were observed by cell morphology and cell viability. Effect of Macrophage Cell Line RAW2 6 4.7 on Oxidative Damage in Cells. Results tb OOH could induce the injury of RAW264 cells, which showed cell shrinkage, disappearance of pseudopodia and decrease of cell viability, and the damage was obvious with the increase of tb OOH concentration; while M-CSF could reduce tb OOH induced RAW2 6 4.7 cell oxidative damage. Conclusion M-CSF can protect the monocyte-macrophage from oxidative damage.
