目的 观察大蒜素对小鼠脾脏指数及其T淋巴细胞亚群分化的影响.方法 将80只清洁级昆明小鼠随机化分组:生理盐水对照组、大蒜素低剂量组、大蒜素中剂量组和大蒜素高剂量组,每组20只,并分别按0.1ml/ (10 gbw)、100 mg/ (kg bw)、200 mg/ (kg bw)、400 mg/ (kg bw)的剂量连续灌胃14d,每天灌胃1次并称重.灌胃结束7d后处死小鼠,无菌分离脾脏称重,计算脾脏系数,制作单细胞悬液,采用流式细胞仪检测小鼠T淋巴细胞亚群中CD4+T淋巴细胞、CD8+T淋巴细胞和CD4+/CD8+的值.结果 大蒜素中、高剂量组和生理盐水对照组比较,脾脏指数、脾脏淋巴细胞CD4+T淋巴细胞比例和CD4+/CD8+值均升高,CD8+T淋巴细胞比例降低,差异有统计学意义(F=5.52,P<0.05),而大蒜素低剂量组与生理盐水对照组比较,脾脏指数、脾脏淋巴细胞CD4+T淋巴细胞、CD8+T淋巴细胞和CD4+/CD8+值差异无统计学意义(F=1.87,P>0.05).结论 大蒜素能够使得小鼠脾脏指数、脾脏淋巴细胞CD4+T淋巴细胞比例和CD4+/CD8+值均升高,CD8+T淋巴细胞比例降低,对正常小鼠脾脏免疫功能具有正向调节作用.“,”Objective To observe the effects of allicin on spleen T lymphocyte subset in mice.Methods Eighty healthy adult Kunming mice were randomly and evenly divided into 4 groups,including normal saline control group,allicin low-dose group,mid-dose group and high-dose group,which were treated with normal saline and 0.1 ml/ (10g bw) of allicin at 100 mg/ (kg bw),200 mg/ (kg bw) and 400 mg/ (kg bw) by gavage once a day for 14 days.They were sacrificed 7 days after the final gavage and the spleens were collected to obtain the spleen index and to make single cell suspension.FCM was used to determine the percentage of CD4+ and CD8+ T lymphocyte and to calculate the CD4+/CD8+ ratio.Results The amount of CD4+ T cells,CD4+/CD8+ ratio and the spleen index were higher in the mid-dose and high-dose allicin mice group compared with normal saline control group,while the amount of CD8+ T cells were lower,the differences were significant (F=5.52,P<0.05).However,the differences were not statistically significant when compared the lowdose group with the control group (F=1.87,P>0.05).Conclusions Mid-dose and high-dose allicin can upregulate CD4+ T cells and down regulate CD8+ T cells and increase the CD4+/CD8+ ratio in the spleen of mice.
