目的:建立芪血通络片中黄芪甲苷的含量测定方法。方法:采用HPLC-ELSD法,色谱柱:Phenomenex Gemini C_(18)110A(250 mm×4.6mm,5μm);流动相:甲醇-水(80:20);流速:1.0 ml·min~(-1);柱温:30℃;进样量:10μl;ELSD检测器漂移管温度:45℃,载气(空气)流速:3.0 L·min~(-1)。结果:黄芪甲苷进样量在11.5~57.5μg范围内,峰面积积分值的对数与进样量的对数线性关系良好,r=0.999 5,平均回收率101.65%,RSD为2.3%。结论:本方法无干扰,准确、重复性良好,可作为含量控制方法。
Objective: To establish a method for the determination of Astragaloside Ⅳ in Qixuetongluo tablets. METHODS: The HPLC-ELSD method was used. The mobile phase consisted of Phenomenex Gemini C 18 18A (250 mm × 4.6 mm, 5 μm), mobile phase methanol-water (80:20), flow rate 1.0 ml · min -1 ); Column temperature: 30 ℃; injection volume: 10μl; ELSD detector drift tube temperature: 45 ℃; carrier gas flow rate: 3.0 L · min -1. Results: Astragaloside injection volume was in the range of 11.5-57.5μg. The logarithm of the peak area integral value was logarithmically linear with the logarithm of injection volume, r = 0.999 5, the average recovery was 101.65% and the RSD was 2.3%. Conclusion: This method has no interference, accurate, good repeatability and can be used as content control method.
