目的:观察高三尖杉酯碱(homoharringtonine,HHT)对视网膜母细胞瘤(retinoblastoma,RB)细胞系HXO-RB44细胞的杀伤作用及其诱导的细胞死亡形式。方法:用MTT法测定肿瘤细胞存活率,提取药物作用后的细胞核DNA进行琼脂糖凝胶电泳。结果:HHT在10-9mol/L~10-4mol/L浓度范围对HXO-RB44具有较强的杀伤作用(P<0.05)。10-6mol/L的HHT与HXO-RB44作用48小时后,核DNA发生有规律的裂解,琼脂糖凝胶电泳呈典型DNA梯度。结论:HHT诱导RB发生程序性死亡(programmedceldeath,PCD),其诱导活性呈较强的浓度和时间依赖性。
OBJECTIVE: To observe the killing effect of homoharringtonine (HHT) on retinoblastoma (RB) cell line HXO-RB44 and its induced cell death pattern. Methods: The survival rate of tumor cells was determined by MTT method. The DNA of the cells after the drug was extracted was subjected to agarose gel electrophoresis. Results: HHT could kill HXO-RB44 in a concentration range of 10-9mol / L ~ 10-4mol / L (P <0.05). Forty-eight hours after treatment with 10-6mol / L HHT and HXO-RB44, the nuclear DNA was cleaved regularly, and the DNA ladder showed a typical DNA gradient on agarose gel electrophoresis. Conclusion: HHT induces programmed programmed death (PCD) and its inducing activity is in a concentration-and time-dependent manner.
